Coronary Reperfusion

Coronary reperfusion is the restoration of blood flow through an obstructed coronary artery, a time-sensitive intervention that limits myocardial ischemia and preserves heart muscle. It can be achieved by dissolving an occluding clot with fibrinolytic drugs or by mechanically reopening the vessel through percutaneous coronary intervention, often with balloon angioplasty and stent placement; restoring oxygen delivery can also trigger inflammation and oxidative stress known as ischemia-reperfusion injury. In medicine, reperfusion is central to treating acute myocardial infarction, improving survival and ventricular function while guiding research into therapies that protect the heart during reoxygenation.

Coronary flow reserve (CFR), is defined as the ratio of maximal coronary blood flow to the resting coronary blood flow. We present a protocol for evaluating CFR in rats via ultrasound, which offers the opportunity to predict cardiovascular risk factors in the absence of obstructive coronary disease.

The present protocol describes a modified parasternal long-axis view for swift and precise localization of the left anterior descending artery. This approach is designed to be simpler and more user-friendly while facilitating the examination of dynamic changes in coronary flow reserve following myocardial ischemia-reperfusion in mice.

Herein, we present a protocol focusing on the quality control of the left anterior descending coronary artery ligation by technically modifying the traditional procedure in rats for acute myocardial ischemia-reperfusion research.

Improved Rodent Model of Myocardial Ischemia and Reperfusion Injury

2022

Myocardial ischemia-reperfusion model of rat heart is improved by using a self-made retractor, polyvinyl chloride tube, and a unique knotting method. Electrocardiogram, triphenyltetrazolium chloride and histological staining, and percent survival analysis results showed that the improved model group has higher success and survival rates than the already existing model group.
