Coronary Vasculature Cast

A coronary vasculature cast is a three-dimensional replica of the arterial and venous network that supplies the heart, providing a detailed view of coronary vessel architecture. To create one, researchers introduce a liquid casting material into the coronary vessels, allow it to solidify within the branching network, and remove the surrounding tissue, leaving a durable mold of the vascular channels. This approach reveals vessel diameter, branching patterns, anastomoses, and spatial relationships that may be difficult to appreciate in conventional sections. Coronary vasculature casts support anatomical education, cardiovascular research, comparative studies, and investigation of vascular remodeling or disease-related changes.

We present a protocol for the analysis of coronary vessels in whole embryonic murine hearts up to E15.5, using standard immunological staining methods followed by optical clearance and confocal microscopy. This technique enables visualization of blood vessels throughout the entire heart without the need for time-consuming analysis of serial sections.

An indirect additive manufacturing method combining a 3D printing of polymers with a centrifugal casting is outlined for manufacturing 3D octet truss metals (Al and Cu alloys) having a unit cell length of 5 mm with a wall thickness of 0.5 mm.
