Differential Expression Analysis

Differential expression analysis is a statistical method for identifying genes whose expression levels differ between biological conditions, such as healthy and diseased tissues, making it valuable for understanding molecular changes in medicine. It compares RNA sequencing or microarray measurements after accounting for factors such as sequencing depth, sample variation, and experimental design, then uses statistical models and multiple-testing correction to distinguish meaningful changes from random noise. Researchers use the results to identify disease-associated pathways, characterize treatment responses, and prioritize potential biomarkers or therapeutic targets. Careful study design and biological validation help determine whether observed expression differences have clinical significance.

We describe a set of assays to analyze expression levels of H1 linker histones. mRNA of individual H1 genes are quantitatively measured by random primer based reverse transcription followed by real-time PCR, whereas protein quantification of H1 histones is achieved by HPLC analysis.
