Ecm Protein Localization

ECM protein localization is the mapping of extracellular matrix proteins within tissues, where their position helps define tissue architecture, cell behavior, and disease-related remodeling. Researchers typically identify proteins such as collagen, fibronectin, or laminin by binding specific antibodies or tags to target molecules, then visualize their distribution with fluorescence or other microscopy methods. In medicine, localization patterns can reveal changes in fibrosis, tumor invasion, wound repair, and vascular disease, linking matrix organization to pathological mechanisms. These analyses support biomarker development, improve interpretation of tissue samples, and guide research on therapies that modify the extracellular environment.

Collagens are the Major Structural Proteins of ECM

2023

Three main types of fibers are secreted by fibroblasts: collagen fibers, elastic fibers, and reticular fibers. Collagen fiber is made from fibrous protein subunits linked together to form a long, straight fiber. Collagen fibers, while flexible, have great tensile strength, resist stretching, and give ligaments and tendons their characteristic resilience and strength. These fibers hold connective tissues together, even during the body's movement. Connective tissue proper includes loose...

Identification of Plasmodesmal Localization Sequences in Proteins In Planta

Plant intercellular connections, the plasmodesmata (Pd), play central roles in plant physiology and plant-virus interactions. Critical to Pd transport are sorting signals that direct proteins to Pd. However, our knowledge about these sequences is still in its infancy. We describe a strategy to identify Pd localization signals in Pd-targeted proteins.

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
