Ecm Ultrastructure Preservation

ECM ultrastructure preservation is the practice of maintaining the native organization of extracellular matrix components, including fibers, pores, and tissue interfaces, during specimen preparation and analysis. It matters because these spatial features influence tissue mechanics, cell behavior, and interpretation of disease-related remodeling. Effective preservation depends on rapid stabilization, compatible fixation, controlled dehydration and embedding, and careful sectioning or imaging to limit shrinkage, distortion, and loss of matrix components. In medicine, preserved ECM ultrastructure supports histopathology, electron microscopy, and research on fibrosis, tumor microenvironments, wound repair, and biomaterial integration, helping link molecular changes to tissue function.

Here we show how to use the sandwich freezing device for rapid freezing of biological specimens, including bacteria, yeast, cultured cells, isolated cells, animal and human tissues, and viruses. We also show how to prepare specimens for ultrathin sectioning after rapid freezing.
