Endothelial Rna Isolation

Endothelial RNA isolation is a laboratory method for extracting high-quality RNA from endothelial cells, which line blood vessels and regulate vascular function, inflammation, and permeability. The process typically involves disrupting cells, rapidly inactivating RNases, separating RNA from DNA and proteins, and purifying it under conditions that limit degradation. The recovered RNA supports gene-expression analysis through methods such as reverse transcription and quantitative PCR, helping researchers investigate angiogenesis, vascular injury, cardiovascular disease, and responses to drugs or inflammatory signals. We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

Primary cell culture is a useful technique for analyzing specific populations of cells, particularly from transgenic mouse embryos at specific developmental stages. Herein, embryonic ventricles are dissected and dissociated, and antibody-conjugated beads recognize and separate out the endothelial cells for further analysis.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

Novel isolation methods of primary endothelial cells from blood vessels are needed. This protocol describes a new technique that completely inverts blood vessels of interest, exposing only the endothelial side to enzymatic digestion. The resulting pure endothelial cell culture can be used to study cardiovascular diseases, disease modelling, and angiogenesis.
