Human Brain Tissue

Human brain tissue is the specialized biological material that forms the brain, where neurons, glial cells, blood vessels, and extracellular structures work together to support sensation, movement, cognition, and vital regulation. Its function depends on electrical impulses and chemical neurotransmission across synapses, while glial cells maintain the cellular environment, provide metabolic support, and help regulate signaling. In medicine, human brain tissue obtained through surgery, biopsy, or postmortem examination supports the diagnosis and study of tumors, traumatic injury, stroke, and neurodegenerative disease. Histological, molecular, and cellular analyses of this tissue can reveal disease mechanisms, guide treatment development, and improve understanding of human neurological disorders.

Until recently, expression studies on human brain were limited to quantification of RNA or protein. With the chromatin immunoprecipitation techniques described in this paper, it will be possible to map histone methylation and other epigenetic regulators of gene expression in postmortem brain.

Source: Eid, L. & Parent, M. Preparation of Non-human Primate Brain Tissue for Pre-embedding Immunohistochemistry and Electron Microscopy. J. Vis. Exp. (2017).This video demonstrates the preparation of aldehyde-fixed non-human primate brain tissue using pre-embedding immunohistochemistry for electron microscopy. It outlines the steps for immunolabeling a specific neuronal protein, followed by a peroxidase reaction that produces a colored precipitate, enabling visualization of the target...

To detect these bacteria, take a deparaffinized, rehydrated, bacteria-infected human brain section with exposed protein antigens and inactivated endogenous phosphatase.

Isolation of Cerebral Capillaries from Human Brain: An Optimized Method to Extract Cerebral Capillaries from Human Brain Tissue

The cellular heterogeneity of brain tissue poses a significant limitation for the study of epigenetic markings in chromatin because most assays lack single cell resolution. Neurons typically are intermingled with glia and other non-neuronal cells. We provide a protocol to extract and collect neuronal nuclei from human brain.
