Neutrophil Elastase Staining

Neutrophil elastase staining is a laboratory method used to detect and localize neutrophil elastase, a proteolytic enzyme stored in the azurophilic granules of neutrophils, within cells or tissue samples. The technique typically uses antibodies that bind specifically to the enzyme, followed by chromogenic or fluorescent detection to reveal its distribution under a microscope; signal intensity and localization can indicate neutrophil presence and activation. In medicine and biomedical research, this staining supports the assessment of acute inflammation, infection, tissue injury, and neutrophil-rich diseases, while helping researchers study immune-cell behavior and evaluate pathological changes in tissue architecture.

This video demonstrates the induction and visualization of neutrophil extracellular traps or NETs in pre-labeled neutrophils using patient serum containing anti-neutrophil cytoplasmic antibodies. Furthermore, using an impermeable DNA dye and fluorescence microscopy enables the visualization of NETs.

Neutrophil Extracellular Traps (NETs) are an important innate immune mechanism to fight pathogenic bacteria, fungi and parasites. Here we describe methods to isolate neutrophil granulocytes from human blood and to activate them to form NETs. We present preparation techniques to visualize NETs in light and electron microscopy.

This video shows how to induce abdominal aortic aneurysms (AAA) in mice via transient intraluminal infusion of porcine pancreatic elastase into the infrarenal segment of the abdominal aorta. The model has the ability to add broad insight into the pathobiology of AAA due to the emergence of numerous transgenic and gene knockout mice.
