Preclinical Transplant Model

A preclinical transplant model is an experimental system used to evaluate transplantation strategies, graft performance, and safety before clinical testing in humans. Typically, researchers place donor cells, tissues, or organs into a recipient organism and monitor how immune recognition, inflammation, vascular integration, and immunosuppressive treatment affect graft survival and function under controlled conditions. These models help identify mechanisms of rejection, optimize surgical and therapeutic protocols, and assess emerging approaches such as tissue engineering or immune tolerance induction. By linking laboratory findings with clinical outcomes, preclinical transplant models support evidence-based development of safer, more effective transplant therapies.

Patient-derived xenograft (PDX) models and transplantable genetically engineered mouse models faithfully recapitulate human disease and are preferred models for basic and translational breast cancer research. Here, a method is described to orthotopically transplant breast tumor fragments into the mammary fat pad to study tumor biology and evaluate drug response.
