Protein Leakage Assay

A protein leakage assay is an analytical method that measures proteins released from cells, tissues, or biological compartments to assess membrane integrity and cellular damage. When a membrane becomes compromised by injury, disease, toxins, or treatment, intracellular proteins escape into the surrounding fluid; the collected sample is then analyzed with a protein-detection method and compared with appropriate controls. In medicine, this assay supports evaluation of cytotoxicity, drug effects, tissue injury, and membrane-disrupting processes. By providing a quantitative indicator of structural damage, protein leakage measurements can help characterize disease mechanisms and assess the safety or therapeutic potential of clinical interventions.

The accuracy and sensitivity of protein determination by the rapid and convenient Bradford assay is compromised by intrinsic nonlinearity. We show a simple linearization procedure that greatly increases the accuracy, improves the sensitivity of the assay about 10-fold, and significantly reduces interference by detergents.
