Rabbit Atherosclerosis Model

The rabbit atherosclerosis model is an experimental disease model used to reproduce and study the development of atherosclerotic plaques, which narrow arteries and contribute to cardiovascular disease. Researchers commonly induce lesions by feeding rabbits cholesterol-rich diets, sometimes combined with arterial injury, causing lipid accumulation, endothelial dysfunction, inflammation, and plaque formation in the vessel wall. Because rabbits develop measurable changes in lipoprotein metabolism and arterial pathology, this model supports investigations of disease mechanisms, imaging methods, preventive strategies, and potential therapies. It also helps researchers evaluate how dietary factors and pharmacological interventions influence plaque growth, composition, and stability before clinical studies.

Animal models of atherosclerosis are essential to understand the mechanism and to investigate newer approaches to prevent plaque development or rupture, a leading cause of death in the industrialized world. This protocol uses a combination of balloon injury and cholesterol rich diet to induce atherosclerotic plaques in rabbit iliac artery.

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Anesthesia is an essential component of rabbit surgical and experimental procedures because it prevents pain, reduces stress responses, and helps immobilize the animal during handling and manipulation when general anesthesia is...

We detail a new near-infrared fluorescence (NIRF) catheter for 2-dimensional intravascular molecular imaging of plaque biology in vivo. The NIRF catheter can visualize key biological processes such as inflammation by reporting on the presence of plaque-avid activatable and targeted NIR fluorochromes. The catheter utilizes clinical engineering and power requirements and is targeted for application in human coronary arteries. The following research study describes a multimodal imaging strategy...
