Sybr Green

SYBR Green is a fluorescent nucleic acid dye used to detect and quantify double-stranded DNA, making it valuable for molecular analysis in medicine and biomedical research. The dye binds to double-stranded DNA and emits substantially stronger fluorescence than in its unbound state, allowing instruments to monitor DNA accumulation during real-time polymerase chain reaction (qPCR) or visualize products after gel electrophoresis. Because SYBR Green binds any double-stranded DNA, assay design and melting-curve analysis help distinguish specific amplification from nonspecific products. Applications include measuring gene expression, detecting infectious agents, identifying genetic targets, and evaluating clinical samples, although probe-based methods may provide greater sequence specificity.

This protocol diagnoses Tilapia Lake Virus (TiLV) in tilapia tissues using RT-PCR methodologies. The entire method is described from tissue dissection to total RNA extraction, followed by cDNA synthesis and detection of TiLV by either conventional PCR or quantitative PCR using dsDNA binding a fluorescent binding dye.
