Tissue Sampling Procedures

Tissue sampling procedures are medical methods used to obtain cells or tissue from the body for diagnostic, prognostic, or research analysis. Depending on the suspected condition and anatomical site, clinicians may collect material through fine-needle aspiration, core-needle biopsy, endoscopic sampling, or surgical excision, using aseptic technique and appropriate guidance to target representative tissue while limiting injury. Samples are then preserved, processed, and examined by histopathology, cytology, molecular testing, or microbiology to identify disease-related changes. These procedures support cancer diagnosis and staging, evaluation of infection and inflammation, treatment planning, and monitoring of therapeutic response.

Quantitation of DNA double-strand breaks on the basis of γH2AX foci has become an invaluable tool, particularly in radiation biology, for the evaluation of tissue radiosensitivity and effects of radiation modifying compounds. Here we demonstrate the use of an immunofluorescence assay for quantitation of γH2AX foci in tissue samples.

Here we demonstrate a protocol to standardize sampling procedures of an established porcine model of acute myocardial infarction in order to increase its translational value in the understanding of the pathophysiology of myocardial ischemia/reperfusion injury and to test novel drug candidates.
