Von Kossa Staining

Von Kossa staining is a histochemical technique used to visualize mineralized deposits, especially calcium-associated phosphate or carbonate salts, in tissue sections, making it valuable for assessing calcification and bone formation. In the procedure, silver ions react with anions in deposited mineral; exposure to light reduces the bound silver to metallic silver, producing dark brown to black areas where mineral is present. In medicine and biomedical research, the method helps identify ectopic calcification, evaluate bone and cartilage mineralization, and assess mineral deposition in cultured cells or engineered tissues. Because it indicates mineral-associated salts rather than calcium directly, results require careful interpretation alongside complementary stains or assays.
