Astrocyte Enrichment

Astrocyte enrichment is a cell-culture strategy used to increase the proportion and relative purity of astrocytes from mixed neural cell preparations, enabling controlled study of these glial cells. It typically combines tissue dissociation with selective culture conditions that favor astrocyte survival or expansion, while differential adhesion, passaging, or removal of other cell types reduces neuronal and oligodendroglial contamination. Enriched astrocyte cultures provide experimental systems for examining neuron–glia communication, inflammatory signaling, neurotoxicity, and responses to injury or disease-associated stimuli. In neuroscience research, the approach supports reproducible cellular assays, disease modeling, and evaluation of compounds that alter astrocyte function.

Astrocytes have been recognized to be versatile cells participating in fundamental biological processes that are essential for normal brain development and function, and central nervous system repair. Here we present a rapid procedure to obtain pure mouse astrocyte cultures to study the biology of this major class of central nervous system cells.

A Simple Technique to Isolate and Culture Murine Astrocytes

The video demonstrates a simple method to culture murine astrocytes in isolation from other brain cells. During culturing, by using astrocyte-specific media and inhibiting the adherence of non-astrocytic cells, the method selectively cultures astrocytes. These cultured astrocytes exhibit stellate morphology with fine processes.
