Brain Endothelial Cells

Brain endothelial cells are specialized cells lining the blood vessels of the central nervous system, where they regulate communication between the bloodstream and neural tissue. Joined by tight junctions and supported by transport proteins, they form the vascular component of the blood-brain barrier, selectively controlling the movement of nutrients, signaling molecules, drugs, and potentially harmful substances. Their interactions with pericytes, astrocytes, and neurons help maintain the brain’s specialized microenvironment. Studying brain endothelial cells supports research into barrier function, neurovascular signaling, inflammation, stroke, neurodegenerative disease, and strategies for delivering therapeutics to the brain.

This method describes how to isolate and immortalize microvascular endothelial cells from mouse brain. We describe a step-by-step protocol starting from the homogenization of brain tissue, digestion steps, seeding and immortalization of the cells. Usually, it takes about five weeks to obtain a homogenous, immortalized microvascular endothelial cell line.

In vitro traumatic brain injury models are being developed to reproduce in vivo brain deformation. Stretch-induced injury has been employed for astrocytes, neurons, glial cells, aortic, and brain endothelial cells. However, our system uses a blood brain barrier (BBB) model that possesses properties constituting a legitimate model of the BBB to establish an in vitro TBI model.
