Craniotomy Procedure

Craniotomy is a neurosurgical procedure in which a temporary section of the skull is removed to provide access to the brain, enabling diagnosis or treatment while preserving the bone for closure. After exposing the skull, the surgeon creates a bone flap, opens the dura mater, and uses microsurgical or image-guided techniques to reach a targeted region while limiting injury to healthy tissue; the flap is then secured after the intervention. In neuroscience, craniotomy supports tumor resection, treatment of vascular abnormalities, evacuation of bleeding, pressure relief, and access for biopsy or research recording, linking clinical care with direct study of brain structure and function.

This video and protocol demonstrate how to implant a glass-covered cranial window in rodents. These preparations can be used for chronic in vivo two-photon imaging of the neocortex over time scales of months. It may also be used for other types of imaging, including optical intrinsic signal imaging.
