Csf Centrifugation Protocol

A CSF centrifugation protocol is a standardized method for processing cerebrospinal fluid by separating its cellular and soluble components, which supports reliable analysis in neuroscience. Centrifugal force drives cells, debris, and other particulate material toward the bottom of a collection tube, producing a supernatant that can be examined separately from the cellular fraction; controlled handling helps preserve sample integrity. This preparation enables cytological assessment, protein and biomarker measurements, and downstream molecular studies of the central nervous system. Consistent centrifugation improves comparability among samples and contributes to research on neuroinflammation, infection, neurodegeneration, and neurological disease.

We designed a cell-free receptor binding assay in order to estimate the binding of granulocyte-macrophage colony-stimulating factor (GM-CSF) to the receptors. It enables us to evaluate competitive inhibition of biotinylated GM-CSF binding to soluble GM-CSF receptor alpha by GM-CSF autoantibody with excellent reproducibility.

Here detailed protocols for culturing the murine myeloid precursor 32D/G-CSF-R cell line, performing viral infections, and carrying out proliferation and differentiation assays are presented.
