Direct Nerve Injection

Direct nerve injection is a neuroscience technique that delivers a solution directly into a peripheral nerve to study or modify neural function. A fine needle penetrates the nerve’s protective connective-tissue layers and deposits a controlled volume within or alongside nerve fascicles, allowing local exposure while limiting distribution to surrounding tissues. Researchers use this approach to investigate axonal transport, nerve regeneration, pain signaling, and the effects of tracers, genes, or pharmacological agents. Because inaccurate placement or excessive pressure can damage nerve fibers, the method requires careful anatomical targeting and controlled injection conditions, making it valuable for both mechanistic studies and the development of localized neural therapies.

Direct Vagus Nerve Injection Protocol For Rats

2025

We present a protocol for direct vagus nerve injection in rats, enabling drug delivery directly into the nerve without post-injection complications. This method applies to preclinical neurological studies involving autonomic nervous system manipulation. It can be used for direct nerve injection for other nerves in rats and other species, with necessary modifications.

Direct injection into the rat optic nerve is useful for regenerative research. We demonstrate a minimally-invasive technique for direct injection into a rat optic nerve that does not involve opening the skull. Using this method, surgical complications are minimized and recovery is more rapid.

Direct Reconnection of Cervical Nerves to Restore Forelimb Mobility in a Mouse

Source: Gao, Z., et al. A Mouse Model of Direct Anastomosis via the Prespinal Route for Crossing Nerve Transfer Surgery. J. Vis. Exp. (2021).This video demonstrates restoring forelimb movement in a mouse via direct cervical nerve reconnection. It begins with a mouse with a paralyzed right forelimb due to a transected right C7 nerve at its nerve root. The left C7 nerve is severed, guided to the right via the prespinal route, and sutured with the right C7 nerve. The surgical site is then cleaned...
