Drg Exposure Procedure

The DRG exposure procedure is a surgical technique used to access a dorsal root ganglion, a cluster of sensory neuron cell bodies that relays information from the peripheral nervous system to the spinal cord. In neuroscience research, controlled removal or separation of overlying tissues and vertebral structures reveals the ganglion while minimizing disruption to the spinal cord, nerve roots, and surrounding anatomy. This access supports electrophysiological recording, targeted drug delivery, imaging, and molecular or histological analysis of sensory neurons. The procedure is particularly valuable for investigating pain, somatosensation, nerve injury, and cellular responses to neurological disease or treatment.

This video demonstrates a procedure for isolating dorsal root ganglion (DRG) neurons from rat embryos and co-culturing them with Schwann cell precursors to generate mature Schwann cells. The isolated DRGs are processed to isolate neurons and glial cells. The isolated cells are cultured to purify the neurons, which are then co-cultured with Schwann cell-like cells (SCLCs) in a co-culture medium. Interaction with the DRG neurons leads to the maturation of SCLCs into Schwann cells.
