Extracellular Stimulation

Extracellular stimulation is the activation or modulation of excitable cells by applying electrical signals outside their membranes, a foundational approach in neuroscience for probing neural function. An external electrode generates an electric field that redistributes charge across the cell membrane, changing membrane potential and potentially driving action potentials when the stimulus reaches threshold; effects depend on amplitude, duration, frequency, and electrode placement. Researchers use extracellular stimulation to map neural circuits, study synaptic and network responses, and control activity in brain, spinal, and peripheral nerves. The same principles support neuroprosthetics and clinical interventions that restore or regulate nervous-system function.

Neutrophil Extracellular Traps (NETs) are an important innate immune mechanism to fight pathogenic bacteria, fungi and parasites. Here we describe methods to isolate neutrophil granulocytes from human blood and to activate them to form NETs. We present preparation techniques to visualize NETs in light and electron microscopy.

The biosynthesis of cartilaginous extracellular matrix by chondrocytes can be affected by application of mechanical stimuli. This method describes the technique of applying dynamic compressive strains to chondrocytes encapsulated in 3D constructs and the evaluation of induced changes in chondrocyte metabolism.
