Glioblastoma Microglia Interaction

Glioblastoma microglia interaction describes the dynamic communication between glioblastoma cells and microglia, the resident immune cells of the central nervous system, within the tumor microenvironment. Glioblastoma cells release signaling molecules that recruit and reprogram microglia, while microglia can adopt tumor-supportive states that promote inflammation, angiogenesis, invasion, and immune suppression. This bidirectional signaling involves changes in cytokine production, phagocytic activity, and gene expression, linking neural immunity with cancer progression. Studying these interactions helps clarify glioblastoma biology and supports the development of therapies that restore microglial antitumor activity, improve immune responses, and potentially limit tumor growth and recurrence.

Understanding the malignant behavior of cancer requires creating accurate models of how tumor cells interact with components of the tumor microenvironment, such as macrophages. Here we describe two methods to study glioblastoma cell interaction with tumor associated macrophages and microglia where the effect on glioblastoma invasion is assessed.

Matrix-Based Tumor Invasion Model Using Microglia Glioblastoma Co-Culture

In this video, glioblastoma and microglia cells are mixed with a polymer matrix solution and transferred to a porous chamber insert where a gel forms, embedding the cells. Microglia stimulate the migration of glioblastoma cells through the porous insert, mimicking tumor cell invasion into surrounding tissue, Cells are then fixed and prepared for further analysis.
