Ipsc Microglia

iPSC microglia are microglia-like immune cells generated from induced pluripotent stem cells (iPSCs), providing a renewable human model for studying the brain’s resident immune system. Researchers reprogram adult somatic cells to pluripotency, then guide them through developmental signals that produce microglial progenitors and mature cells with characteristic immune and phagocytic functions. In neuroscience, these cells can be combined with neurons, astrocytes, or brain organoids to examine neuroinflammation, synaptic interactions, and disease-associated responses in conditions such as Alzheimer’s disease and Parkinson’s disease. Because iPSC lines can come from individual donors, they also support patient-specific disease modeling, therapeutic screening, and studies of genetic influences on microglial biology.

2026

We describe a sensitive, time-resolved quantitative platform to measure clearance of extracellular tau in human induced pluripotent stem cell (iPSC)-derived microglia using a microplate-based luminescence system. This approach enables sensitive detection of tau degradation kinetics in a microwell format, allowing for high-throughput interrogation of tau handling under various experimental conditions.

This video demonstrates a protocol for generating functional antigen-specific T cells from iPSC-derived immature T cells using a three-dimensional thymic culture system. Co-culturing the iPSC-derived immature T cells with endogenous lymphocyte-depleted mice fetal thymic lobes resulted in successful maturation of the T cells to form CD8αβ+ MHC class I+ iPSC-derived thymic emigrants (iTEs).

Small-scale Propagation of Human iPSCs in Serum-free Conditions for Routine Immunocytochemical Characterization

2017

Regular characterization of induced pluripotent stem cells (iPSCs), to ascertain maintenance of their pluripotent state, is an important step before these cells are used for other applications. Here we describe a method for the small-scale propagation of human iPSCs specifically designed to enable their easy and routine characterization via immunocytochemistry.
