Morphological Staining

Morphological staining is a set of laboratory methods that uses dyes or labels to reveal the size, shape, arrangement, and structural features of cells and tissues, making microscopic anatomy visible. In neuroscience, samples are typically fixed to preserve architecture, treated to allow stain penetration, and exposed to reagents that selectively bind cellular components or fill individual neurons, producing contrast under light or fluorescence microscopy. These methods support identification of neuronal cell bodies, dendrites, axons, nuclei, and tissue organization, helping researchers compare normal and diseased anatomy, map neural circuits, and evaluate structural changes after development, injury, or experimental treatment.

This paper describes the procedures for tactile stimulation of rat pups and subsequent Golgi-Cox staining of neuronal morphology. Tactile stimulation is a positive experience that is administered in the perinatal period by stroking pups with a household duster. Golgi-cox staining is a reliable procedure permitting the visualization of entire neurons.

Histological Staining of Neural Tissue

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue.
