Neuron Isolation

Neuron isolation is a laboratory technique used to separate neurons from nervous tissue so they can be studied in a controlled environment. The process typically combines tissue dissection with enzymatic digestion and gentle mechanical dissociation to release individual cells while preserving membrane integrity, viability, and neuronal characteristics. Isolated neurons can then be maintained in culture, characterized by morphology or molecular markers, and examined through imaging, electrophysiology, or molecular assays. In neuroscience research, this approach supports studies of neuronal development, synaptic function, neurotoxicity, disease mechanisms, and responses to experimental treatments.

This study presents a technique for the isolation of neurons from WT neonatal mice. It requires the careful dissection of the spinal cord from the neonatal mouse, followed by the separation of neurons from the spinal cord tissue through mechanical and enzymatic cleavage.

Neuronal Nuclei Isolation from Human Postmortem Brain Tissue

The cellular heterogeneity of brain tissue poses a significant limitation for the study of epigenetic markings in chromatin because most assays lack single cell resolution. Neurons typically are intermingled with glia and other non-neuronal cells. We provide a protocol to extract and collect neuronal nuclei from human brain.
