Perineuronal Nets

Perineuronal nets are specialized extracellular matrix structures that form lattice-like coats around the cell bodies and proximal dendrites of select neurons, particularly fast-spiking inhibitory interneurons. Built from hyaluronan, chondroitin sulfate proteoglycans, link proteins, and tenascin-R, they assemble through interactions with the neuronal surface and restrict the movement and remodeling of nearby synaptic connections. By stabilizing synapses and regulating access to growth-related molecules, perineuronal nets help close developmental critical periods while preserving mature circuit function. Neuroscience research examines their roles in learning, memory, sensory plasticity, aging, and neurological disease, including whether modifying them can restore plasticity after injury or pathological change.

In the following protocol, we describe a very simple way to isolate Neutrophil Extracellular traps (NETs) from human whole blood using readily available reagents. We then demonstrate how the isolated NETs can be used in an in vitro adhesion assay with cancer cells.

In this video, we demonstrate an assay utilizing fluorescence microscopy to detect the adhesion of cancer cells to neutrophil extracellular traps (NETs). In the test wells, there is a notable increase in the number of fluorescently labeled cancer cells adhering to NETs, whereas wells treated with DNase exhibit reduced cancer cell adhesion to NETs.
