Progenitor Cell Isolation

Progenitor cell isolation is the process of separating developmentally immature, self-renewing cells from tissue or mixed cell populations for study and culture. In neuroscience, researchers typically dissociate neural tissue mechanically or enzymatically, then enrich progenitor cells using selective culture conditions, cell-surface markers, or techniques such as fluorescence-activated cell sorting. Isolated neural progenitors can be expanded, differentiated into neuronal or glial lineages, and analyzed for developmental behavior, signaling, and responses to experimental treatments. Reliable isolation produces defined cell populations that support research on neurogenesis, nervous system development, neurological disease mechanisms, and potential cell-based repair strategies.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.

Endothelial colony forming progenitor cells (ECFCs) are a promising tool to study vascular homeostasis and repair.1,2 This paper introduces a novel animal-serum free method for isolation and expansion of ECFC from heparinised adult human peripheral blood with pooled human platelet lysate (pHPL) diminishing the risk of anti-bovine immunisation.

Here we describe an optimized, highly reproducible protocol to isolate Mesodermal Progenitor Cells (MPCs) from human bone marrow (hBM). MPCs were characterized by flow cytometry and nestin expression. They showed the ability to give rise to exponentially growing MSC-like cell cultures while retaining their angiogenic potential.
