Protein Expression Analysis

Protein expression analysis is the measurement of which proteins cells produce and in what amounts, providing molecular insight into cell state and function. In neuroscience, researchers assess protein abundance and distribution in brain tissue or cultured neurons using methods such as immunoblotting, immunostaining, enzyme-linked assays, and mass spectrometry. These approaches rely on selective antibody binding or peptide detection, followed by comparison with controls to quantify target proteins across conditions. The resulting profiles help characterize synaptic signaling, neuronal development, neurodegenerative disease, and responses to drugs or injury while revealing changes in pathways that may guide mechanistic studies and biomarker development.

This video demonstrates the methodology for performing Reverse Phase Protein Arrays (RPPA) to study protein expression patterns from cell lysate. In RPPA, the lysate containing a mixture of proteins is printed on nitrocellulose slides and proteins of interest are analyzed using fluorescently labeled antibodies.

This protocol presents a method to perform quantitative, single-cell in situ analysis of protein expression to study lineage specification in mouse preimplantation embryos. The procedures necessary for collection of blastocysts, whole-mount immunofluorescent detection of proteins, imaging of samples on a confocal microscope, and nuclear segmentation and image analysis are described.

Expression of malarial proteins in cell based systems remains challenging. We demonstrate two step and one step IVT (in vitro translation) cell free expression systems for expressing malarial recombinant rhoptry proteins from HeLa cells. We use a Ni-resin affinity based purification system to purify the rhoptry proteins.
