Protein Subcellular Localization

Protein subcellular localization is the distribution of proteins within specific cellular compartments, a fundamental property that helps determine how cells function. Proteins reach destinations such as the nucleus, mitochondria, synapses, or membranes through targeting signals, molecular transport pathways, and regulated trafficking, while localization can change in response to cellular conditions. In neuroscience, mapping where proteins reside in neurons clarifies processes including neurotransmission, axonal transport, synaptic plasticity, and neuronal signaling. Experimental approaches such as fluorescence microscopy, cell fractionation, and tagged-protein analysis can reveal abnormal localization associated with neurodevelopmental disorders, neurodegeneration, and impaired neuronal communication.

An Immunolabeling Technique to Visualize the Subcellular Localization of Proteins

This video demonstrates a technique for the immunolabeling of sigma-1 receptors in a murine heart tissue section using quantum dots. The heart tissue is fixed, osmicated, stained, dehydrated, embedded in resin, and then sectioned. The sections are then treated with antibodies and labeled with quantum dots to demarcate the subcellular localization of Sigma-1 receptors using electron microscopy.

Labeling the extracellular domain of a membrane protein with a pH sensitive fluorophore, superecliptic pHluorin (SEP), allows subcellular localization, expression, and trafficking to be determined. Imaging SEP-labeled proteins with total internal reflection fluorescence microscopy (TIRFM) enables the quantification of protein levels in the peripheral ER and plasma membrane.

Nano-fEM: Protein Localization Using Photo-activated Localization Microscopy and Electron Microscopy

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.

This video demonstrates a method for monitoring infected cell protein 0, ICP0 trafficking in herpes simplex virus-1 infection. Post-de novo synthesis, ICP0 translocates to the nucleus, later moving to the cytoplasm during infection progression. Immunofluorescence microscopy reveals and analyzes the protein's subcellular localization, offering insights into its trafficking across infection phases.
