Psoas Muscle Dissection

Psoas muscle dissection is a methodical anatomical examination of the psoas major and its surrounding structures, providing a clear view of relationships that are important in movement, pain, and neural function. During dissection, tissue is removed layer by layer to trace the muscle from its lumbar vertebral attachments toward the lesser trochanter while identifying the lumbar plexus, which forms within the muscle and gives rise to major peripheral nerves. In neuroscience education and research, this approach connects musculoskeletal anatomy with nerve organization, supports interpretation of neurological deficits and regional pain, and strengthens understanding of how deep anatomical structures guide clinical examination and surgical planning.

We present a protocol for dissection of fluorescent-labeled flight muscle from live pupae to generate highly enriched samples ideal for proteomics and deep-sequencing. These samples can offer important mechanistic insights into diverse aspects of muscle development.
