Pupal Retina Staining

Pupal retina staining is a histological and immunolabeling technique used to visualize cells, proteins, and structural features in the developing retina during the pupal stage, a key period of neural circuit formation. The method typically preserves and permeabilizes pupal eye tissue, uses antibodies or fluorescent dyes to bind target molecules, and applies microscopy to reveal their location and organization. In neuroscience, this approach helps researchers track photoreceptor differentiation, neuronal arrangement, synaptic development, and changes caused by genetic or environmental perturbations. By linking molecular patterns with retinal structure, pupal retina staining supports studies of visual system development, neurobiology, and mechanisms of neural connectivity.

The Drosophila retina is a crystal-like lattice composed of a small number of cell types that are generated in a stereotyped manner 1. Its amenability to sophisticated genetic analysis allows the study of complex developmental programs. This protocol describes dissections and immunohistochemistry of retinas at three discrete developmental stages, with a focus on photoreceptor differentiation.

Whole Mount Immunofluorescent Staining of the Neonatal Mouse Retina to Investigate Angiogenesis In vivo

The neonatal murine retina provides a well characterized physiological model of angiogenesis, which permits investigations of the roles of different genes or drugs that modulate angiogenesis in an in vivo context. Immunofluorescent staining to accurately visualize the vascular plexus is pivotal to the success of these types of studies.

Dissection and Staining of Drosophila Pupal Ovaries

The Drosophila ovary is an excellent model system for studying stem cell niche development. Though methods for dissecting larval and adult ovaries have been published, pupal ovary dissections require different techniques that have not been published in detail. Here we outline a protocol for dissecting, staining, and mounting pupal ovaries.

Insect Puparium Removal: A Method for Pupal Investigation

2023

This article describes a technique to remove an insect’s puparium while keeping the inside pupa intact and therefore fit for further investigation. The example protocol shows the procedure with Drosophila guttifera; however, this technique is applicable to other Drosophila species.
