Secretome Analysis

Secretome analysis is the systematic study of proteins, peptides, metabolites, and other molecules released by cells into their surrounding extracellular environment, providing insight into cell communication and function. In neuroscience, researchers collect conditioned media, remove cells and debris, enrich secreted components, and identify or quantify them using proteomics, biochemical assays, or complementary analytical methods. This approach helps characterize signals exchanged by neurons, glial cells, and neural stem cells, including factors involved in synaptic regulation, neuroinflammation, and tissue repair. Secretome profiles can reveal disease-associated biomarkers, clarify mechanisms of cell interaction, and support development of cell-based therapies and regenerative strategies.

Human Adipose Tissue Micro-fragmentation for Cell Phenotyping and Secretome Characterization

Here, we present human adipose tissue enzyme-free micro-fragmentation using a closed system device. This new method allows the obtainment of sub-millimeter clusters of adipose tissue suitable for in vivo transplantation, in vitro culture, and further cell isolation and characterization.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.

Salmonella invades and replicates inside intestinal epithelial cells both in Salmonella-specific vacuoles and free in the cytosol (hyper-replication). A high-throughput fluorescence microscopy-based protocol is described here to quantify the intracellular phenotypes of Salmonella by two complementary image analyses through ImageJ, reaching single-cell resolution and scoring.

A Hydroponic Co-cultivation System for Simultaneous and Systematic Analysis of Plant/Microbe Molecular Interactions and Signaling

The described hydroponic cocultivation system supports intact plants with metal mesh screens and cocultivates them with bacteria. Plant tissue, bacteria, and secreted molecules can then be separately harvested for downstream analyses, simultaneously allowing for the molecular responses of both plant hosts and interacting microbes or microbiomes to be investigated.

The combination of iterative-bleaching-extends-multiplexity (IBEX) and a commercial nucleotide labeling assay (Click-iT EdU) enables the detection and categorization of dividing cell types in highly dynamic processes in fixed frozen murine tissue sections. Furthermore, a novel open-source image processing pipeline provides high-throughput image acquisition and analysis.
