Activated Oxygen Complex

An activated oxygen complex is a reactive oxygen-containing intermediate that enables difficult oxidation reactions, making it important for understanding how drugs are transformed in the body. In pharmacology, enzymes such as cytochrome P450 bind molecular oxygen and use electrons and protons to generate a highly reactive iron-oxygen species, which transfers an oxygen atom to a drug or other substrate. These reactions produce metabolites with altered activity, solubility, and elimination profiles. Studying activated oxygen complexes helps explain drug metabolism, pharmacokinetic variability, toxic metabolite formation, and potential drug interactions, while also supporting the design of safer and more effective therapeutics.

This video demonstrates a protocol for assessing the activation of dendritic cells using immunoglobulin G (IgG)-coated tumor cells. Dendritic cells internalize the IgG-coated tumor cells, process the tumor antigens, and present them on the surface through MHC-II molecules, along with the co-expression of the costimulatory molecule CD86. These dendritic cells then undergo MHC-II and CD86-targeted immunostaining and flow cytometry analysis to identify their activation state.

Here, we present a protocol to assess physical activity intensity levels measured with indirect calorimetry and two accelerometers (on the right wrist and waist) during an incremental walking-jogging test (from 0.84 to 2.37 m/s) on an oval track.
