Hemispherical Drops

Hemispherical drops are liquid droplets shaped like half-spheres, typically resting on a flat surface, and they provide a simple model for studying surface tension and wetting in physics. Their form results from a balance between cohesive forces within the liquid, adhesive forces at the solid-liquid interface, and gravity; a contact angle near 90° produces an approximately hemispherical profile. Analyzing the drop’s curvature, volume, and contact angle helps characterize surface wettability and interfacial energy. Hemispherical drops are therefore useful in demonstrations and measurements involving capillarity, droplet formation, material surfaces, and the mechanics of liquid interfaces.

This protocol describes an improved explant procedure that involves ex utero electroporation, dissection and culture of entire cerebral hemispheres from the embryonic mouse. The preparation facilitates pharmacological studies and assays of gene function during early cortical development.

This video demonstrates in vivo recording of local field potential (LFP) from bilateral M2 areas of the mouse brain. An anesthetized mouse is secured and prepared for the procedure. To record LFPs, insert microelectrodes into the target coordinates of the bilateral M2 areas, which are responsible for complex motor movements.

Repetitive Transcranial Magnetic Stimulation to the Unilateral Hemisphere of Rat Brain

We applied repetitive transcranial magnetic stimulation (rTMS) to the unilateral hemisphere of rat brain, by placing a 25-mm figure-8 coil 1 cm lateral to the vertex on the biauricular line and angulating the coil by 45°. An in-house water cooling system was used for rTMS for more than 20 min.

We have developed a technique for picoinjecting microfluidic drops that does not require metal electrodes. As such, devices incorporating our technique are simpler to fabricate and to use.
