Wavelength Determination

Wavelength determination is the measurement of the spatial period of a wave, a fundamental quantity that characterizes electromagnetic radiation, sound, and matter waves. In physics, it commonly uses the relationship λ = v/f, where wavelength equals wave speed divided by frequency, or analyzes interference and diffraction patterns in which wave spacing produces measurable maxima and minima. Accurate wavelength measurements support spectroscopy, optical instrument calibration, communication technology, and the identification of materials through their characteristic spectral lines. The method also connects observable wave behavior with properties such as frequency, energy, and propagation speed.

The technique presented here measures the path of freely swimming microscopic species using single wavelength exposure. C. elegans are used to demonstrate shadow imaging as an inexpensive alternative to costly microscopes. This technique can be adapted to accommodate various orientations, environments and species to measure direction, speed, acceleration and forces.
