Transposable elements (TEs) are proposed as a basis for developing drive systems to spread pathogen resistance genes through vector mosquito populations. The use of transcriptional and translational control DNA elements from genes expressed specifically in the insect germ line to mediate transposition offers possibilities for mitigating some of the concerns about transgene behavior in the target vector species and eliminating effects on nontarget organisms. Here, we describe the successful use of the promoter and untranslated regions from the nanos (nos) orthologous gene of the yellow fever mosquito, Aedes aegypti, to control sex- and tissue-specific expression of exogenously derived mariner MosI transposase-encoding DNA. Transgenic mosquitoes expressed transposase mRNA in abundance near or equal to the endogenous nos transcript and exclusively in the female germ cells. In addition, MosI mRNA was deposited in developing oocytes and localized and maintained at the posterior pole during early embryonic development. Importantly, four of five transgenic lines examined were capable of mobilizing a second MosI transgene into the mosquito genome, indicating that functional transposase was being produced. Thus, the nos control sequences show promise as part of a TE-based gene drive system.

Die Verwendung von transkriptionelle und translational Control DNA-Elemente von Genen, die speziell in die Insekt Keimbahn Umsetzung vermitteln ausgedr&uuml;ckt bietet M&ouml;glichkeiten f&uuml;r Schadensbegrenzende einige Bedenken &uuml;ber Transgene Verhalten in der Vektor-Zielarten und Effekte auf nontarget Organismen zu beseitigen. Hier beschreiben wir den erfolgreichen Einsatz des Projekttr&auml;gers und un&uuml;bersetzte Regionen aus der Nanos (Nos) Orthologous gen der Gelbfieber-M&uuml;cke Aedes Aegypti, zu steuernder Geschlecht und Gewebe-spezifischen Ausdruck abgeleitet exogen Mariner MosI Transposase-Kodierung DNA. Transgene M&uuml;cken ausgedr&uuml;ckt Transposase mRNA in H&uuml;lle und F&uuml;lle in der N&auml;he von oder gleich auf die endogene Nos-Abschrift und ausschlie&szlig;lich in den weiblichen Keimzellen. Dar&uuml;ber hinaus war MosI mRNA hinterlegt bei der Entwicklung von Eizellen und lokalisiert und am hinteren Pol w&auml;hrend der fr&uuml;hen Embryonalentwicklung gepflegt. Wichtig ist, waren vier der f&uuml;nf transgene Linien untersucht Lage Mobilisierung einer zweiten MosI-Transgen in das Moskito-Genom, das angibt, dass die funktionale Transposase produziert wurde. Los elementos transponibles (ET), se proponen como base para el desarrollo de sistemas de transmisi&#xF3;n para difundir los genes de pat&#xF3;genos a trav&#xE9;s de la resistencia a las poblaciones de mosquitos vectores. El uso de elementos de ADN de control de la transcripci&#xF3;n y traducci&#xF3;n de genes que se expresan espec&#xED;ficamente en la l&#xED;nea germinal del insecto para mediar en la transposici&#xF3;n ofrece la posibilidad de mitigar algunas de las preocupaciones sobre el comportamiento de los transgenes en las especies objetivo de vectores y la eliminaci&#xF3;n de los efectos en organismos no atacados. A continuaci&#xF3;n, describimos el uso exitoso de las regiones promotoras y no traducida de los nanos (NOS) de genes ort&#xF3;logos del mosquito de la fiebre amarilla, Aedes aegypti, para controlar el sexo y la expresi&#xF3;n espec&#xED;fica de tejido de forma ex&#xF3;gena marino derivado de Mosi-ADN que codifica la transposasa. Los mosquitos transg&#xE9;nicos expresan ARNm transposasa en abundancia cerca o igual a la transcripci&#xF3;n de los n&#xFA;meros end&#xF3;geno y exclusivamente en las c&#xE9;lulas germinales femeninas. Adem&#xE1;s, el ARNm MoSI fue depositado en los ovocitos de desarrollo y localizados y se mantiene en el polo posterior durante el desarrollo embrionario temprano. Es importante destacar que cuatro de las cinco l&#xED;neas transg&#xE9;nicas analizadas fueron capaces de movilizar a un segundo transgen MoSI en el genoma del mosquito, lo que indica que transposasa funcional se est&#xE1; produciendo. Aedes aegypti mosquitoes are the main vectors of dengue viruses to humans. Understanding their biology and interactions with the pathogen are prerequisites for development of dengue transmission control strategies. Mosquito salivary glands are organs involved directly in pathogen transmission to vertebrate hosts. Information on the spatial distribution of gene expression in these organs is expected to assist in the development of novel disease control strategies, including those that entail the release of transgenic mosquitoes with impaired vector competence.

Mosquito eggshells show remarkable diversity in physical properties and structure consistent with adaptations to the wide variety of environments exploited by these insects. We applied proteomic, transcriptomic, and hybridization in situ techniques to identify gene products and pathways that participate in the assembly of the Aedes aegypti eggshell. Aedes aegypti population density is low during cold and dry seasons and increases immediately after rainfall. The survival of embryos through unfavorable periods is a key factor in the persistence of their populations. The work described here supports integrated vector control approaches that target eggshell formation and result in Ae. aegypti drought-intolerant phenotypes for public health initiatives directed to reduce mosquito-borne diseases.
