The mitochondrial theory of aging proposes that reactive oxygen species (ROS) generated inside the cell will lead, with time, to increasing amounts of oxidative damage to various cell components. The main site for ROS production is the respiratory chain inside the mitochondria and accumulation of mtDNA mutations, and impaired respiratory chain function have been associated with degenerative diseases and aging. The theory predicts that impaired respiratory chain function will augment ROS production and thereby increase the rate of mtDNA mutation accumulation, which, in turn, will further compromise respiratory chain function. Previously, we reported that mice expressing an error-prone version of the catalytic subunit of mtDNA polymerase accumulate a substantial burden of somatic mtDNA mutations, associated with premature aging phenotypes and reduced lifespan. Here we show that these mtDNA mutator mice accumulate mtDNA mutations in an approximately linear manner. The amount of ROS produced was normal, and no increased sensitivity to oxidative stress-induced cell death was observed in mouse embryonic fibroblasts from mtDNA mutator mice, despite the presence of a severe respiratory chain dysfunction. Expression levels of antioxidant defense enzymes, protein carbonylation levels, and aconitase enzyme activity measurements indicated no or only minor oxidative stress in tissues from mtDNA mutator mice. The premature aging phenotypes in mtDNA mutator mice are thus not generated by a vicious cycle of massively increased oxidative stress accompanied by exponential accumulation of mtDNA mutations. We propose instead that respiratory chain dysfunction per se is the primary inducer of premature aging in mtDNA mutator mice.

Somatic mtDNA mutations cause aging phenotypes without affecting reactive oxygen species production.

La th&eacute;orie mitochondriale du vieillissement propose que les esp&egrave;ces oxyg&eacute;n&eacute;es radicalaires (ROS) produites &agrave; l&#39;int&eacute;rieur de la cellule dirigera, avec le temps, de plus en plus de dommages oxydatifs aux diverses composantes de la cellule. Le site principal pour la production de ROS est la cha&icirc;ne respiratoire &agrave; l&#39;int&eacute;rieur de la mitochondrie et l&#39;accumulation de mutations de l&#39;ADNmt et fonction de la cha&icirc;ne respiratoire alt&eacute;r&eacute;e ont &eacute;t&eacute; associ&eacute;s &agrave; des maladies d&eacute;g&eacute;n&eacute;ratives et le vieillissement. La th&eacute;orie pr&eacute;dit que fonction de la cha&icirc;ne respiratoire alt&eacute;r&eacute;e va augmenter la production de ROS et ainsi augmenter le taux d&#39;accumulation de mutations ADNmt, qui, &agrave; son tour, compromet davantage fonction de la cha&icirc;ne respiratoire. Auparavant, nous a signal&eacute; que des souris exprimant une version sujette de la sous-unit&eacute; catalytique de la polym&eacute;rase de l&#39;ADN mitochondrial accumulent un lourd fardeau de mutations de l&#39;ADNmt somatique, associ&eacute;e aux ph&eacute;notypes de vieillissement pr&eacute;matur&eacute; et r&eacute;duit la dur&eacute;e de vie. Ici, nous montrons que ces souris mutantes de mtDNA accumulent des mutations de l&#39;ADNmt d&#39;une mani&egrave;re approximativement lin&eacute;aire. Le montant de ROS produites &eacute;tait normal, et pas une sensibilit&eacute; accrue &agrave; la mort cellulaire d&#39;induite par le stress oxydatif a &eacute;t&eacute; observ&eacute;e dans les fibroblastes embryonnaires de souris de souris de mutateur ADNmt, malgr&eacute; la pr&eacute;sence d&#39;un dysfonctionnement de la cha&icirc;ne respiratoire s&eacute;v&egrave;re. Niveaux d&#39;expression des antioxydants de d&eacute;fense enzymes, prot&eacute;ines carbonylation niveaux et aconitase enzyme activit&eacute; mesures non indiqu&eacute;e ou mineures stress oxydatif dans les tissus des souris mutantes ADNmt. Les ph&eacute;notypes de vieillissement pr&eacute;matur&eacute; chez les souris mutantes ADNmt ne sont donc pas g&eacute;n&eacute;r&eacute;s par un cercle vicieux de massivement une augmentation du stress oxydatif accompagn&eacute; d&#39;accumulation exponentielle des mutations de l&#39;ADNmt. Au lieu de cela, nous vous proposons que ce dysfonctionnement de la cha&icirc;ne respiratoire est en soi l&#39;inducteur principal du vieillissement pr&eacute;matur&eacute; chez les souris mutantes ADNmt.

Des mutations somatiques ADNmt provoquent des ph&eacute;notypes de vieillissement sans affecter la production d&#39;esp&egrave;ces r&eacute;actives de l&#39;oxyg&egrave;ne.

Die mitochondriale Theorie des Alterns schl&auml;gt vor, dass reaktive Sauerstoffspezies (ROS), die innerhalb der Zelle erzeugt mit der Zeit zunehmenden Mengen von oxidativen Sch&auml;den an verschiedenen Zellbestandteile f&uuml;hrt. Die Hauptseite f&uuml;r ROS-Produktion ist die Atmungskette in den Mitochondrien und Anh&auml;ufung von MtDNA-Mutationen und gest&ouml;rte Atmungskette-Funktion wurden mit degenerativen Erkrankungen und Alterung. Die Theorie sagt voraus, dass gest&ouml;rte Atmungskette Funktion ROS-Produktion erg&auml;nzen und damit zu einer Erh&ouml;hung die Rate der MtDNA Mutation Akkumulation, die wiederum weitere Atmungskette Funktion beeintr&auml;chtigt wird. Zuvor wir berichtet, dass M&auml;use, die Ausdruck einer fehlerbehaftete Version die katalytische Untereinheit der MtDNA-Polymerase eine erhebliche Belastung der somatischen MtDNA-Mutationen ansammeln, mit vorzeitiger Alterung Ph&auml;notypen und reduziert die Lebensdauer. Hier zeigen wir, dass diese MtDNA Mutator M&auml;use MtDNA-Mutationen in etwa lineare Weise sammeln. Die Menge der produzierten ROS war normal, und keine erh&ouml;hter Empfindlichkeit gegen&uuml;ber oxidativem Stress-induced Zellentod in M&auml;useembryonalen MtDNA Mutator M&auml;use, trotz des Vorhandenseins von einer schweren Atmungskette Dysfunktion festgestellt. Stufen des Antioxidans Verteidigung Enzyme, Protein Carbonylierung Ebenen und Aconitase Enzym-Aktivit&auml;t Messungen angegebenen keine oder nur geringf&uuml;gige oxidativer Stress in Geweben von MtDNA Mutator M&auml;usen. Die vorzeitige Alterung Ph&auml;notypen MtDNA Mutator M&auml;usen werden somit nicht von einem Teufelskreis der massiv erh&ouml;hten oxidativen Stress, begleitet von exponentielle Anreicherung von MtDNA-Mutationen generiert. Stattdessen schlagen wir vor, dass die Atmungskette Dysfunktion per se ist die prim&auml;re Induktor vorzeitiger Alterung MtDNA Mutator M&auml;usen.

Somatische MtDNA-Mutationen verursachen Alterung Ph&auml;notypen ohne reaktive Sauerstoff Spezies.

La teoria mitocondriale dell&#39;invecchiamento propone che specie reattive dell&#39;ossigeno (ROS) generate all&#39;interno della cellula porter&agrave;, con il tempo, ad aumentare la quantit&agrave; di danno ossidativo a varie componenti cellulari. Il sito principale per la produzione di ROS &egrave; la catena respiratoria all&#39;interno dei mitocondri e l&#39;accumulo di mutazioni del mtDNA, e catena respiratoria alterata funzione sono stati associati con malattie degenerative e l&#39;invecchiamento. La teoria prevede che la catena respiratoria alterata funzione sar&agrave; aumentare la produzione di ROS e quindi aumentare il tasso di accumulo di mutazioni del mtDNA, che, a sua volta, sar&agrave; ulteriormente compromettere la funzione della catena respiratoria. In precedenza, ha riferito che topi che esprimono una error-prona versione della subunit&agrave; catalitica della polimerasi del DNA mitocondriale di accumulano un notevole onere di mutazioni somatiche del DNA mitocondriale, associato a fenotipi di invecchiamento precoce e ridotto la durata della vita. Qui vi mostriamo che questi topi di mtDNA mutator accumulano mutazioni del mtDNA in modo approssimativamente lineare. La quantit&agrave; di ROS prodotte era normale, e non aumenta la sensibilit&agrave; alla morte cellulare indotta da stress ossidativo &egrave; stata osservata in fibroblasti embrionali del mouse da topi di mtDNA mutator, nonostante la presenza di una disfunzione della catena respiratoria severa. I livelli di espressione di antiossidanti di difesa degli enzimi, proteine carbonilazione livelli e aconitasi enzima attivit&agrave; misurazioni non indicato o solo minor stress ossidativo nei tessuti da topi di mtDNA mutator. Cos&igrave; i fenotipi di invecchiamento precoce nei topi di mtDNA mutator non vengono generati da un circolo vizioso di massicciamente aumentato stress ossidativo accompagnati da esponenziale accumulo di mutazioni del mtDNA. Proponiamo invece che tale disfunzione della catena respiratoria per s&eacute; &egrave; l&#39;induttore primario di invecchiamento precoce nei topi di mtDNA mutator.

La teor&iacute;a mitocondrial del envejecimiento propone que dar&aacute; lugar especies reactivas del ox&iacute;geno (ROS) generadas dentro de la c&eacute;lula, con el tiempo, a cantidades crecientes de da&ntilde;o oxidativo en varios componentes de la c&eacute;lula. El sitio principal para la producci&oacute;n de ROS es la cadena respiratoria dentro de la mitocondria y la acumulaci&oacute;n de mutaciones del mtDNA, y funci&oacute;n de la cadena respiratoria deteriorada se han asociado con enfermedades degenerativas y el envejecimiento. La teor&iacute;a predice que la cadena respiratoria deteriorada funci&oacute;n aumentar la producci&oacute;n de ROS y consiguiente aumento de la tasa de acumulaci&oacute;n de mutaci&oacute;n del mtDNA, que, a su vez, m&aacute;s afectar&aacute;n la funci&oacute;n de la cadena respiratoria. Previamente, nos inform&oacute; que los ratones que expresaban una versi&oacute;n propensa de la subunidad catal&iacute;tica de la polimerasa de mtDNA acumulan una importante carga de mutaciones som&aacute;ticas del mtDNA, asociados con fenotipos de envejecimiento prematuro y reduce la vida &uacute;til. Aqu&iacute; mostramos que estos ratones de mutator mtDNA acumulan las mutaciones del mtDNA en forma aproximadamente lineal. La cantidad de ROS producido era normal, y no se observ&oacute; ning&uacute;n aumento de la sensibilidad a la muerte celular inducida por el estr&eacute;s oxidativo en fibroblastos embrionarios de rat&oacute;n de ratones de mtDNA mutator, a pesar de la presencia de una disfunci&oacute;n de la cadena respiratoria severa. Los fenotipos de envejecimiento prematuro en ratones de mtDNA mutator as&iacute; no se generan por un c&iacute;rculo vicioso de masivamente aumento del estr&eacute;s oxidativo acompa&ntilde;ado por acumulaci&oacute;n exponencial de las mutaciones del mtDNA. Proponemos en cambio que la disfunci&oacute;n respiratoria cadena per se es el principal inductor de envejecimiento prematuro en ratones de mtDNA mutator.

Las mutaciones del mtDNA som&aacute;tica provocar fenotipos de envejecimiento sin afectar la producci&oacute;n de especies reactivas de ox&iacute;geno.

Induced pluripotent stem cells (iPSCs) outwardly appear to be indistinguishable from embryonic stem cells (ESCs). A study of gene expression profiles of mouse and human ESCs and iPSCs suggests that, while iPSCs are quite similar to their embryonic counterparts, a recurrent gene expression signature appears in iPSCs regardless of their origin or the method by which they were generated. Upon extended culture, hiPSCs adopt a gene expression profile more similar to hESCs; however, they still retain a gene expression signature unique from hESCs that extends to miRNA expression. Genome-wide data suggested that the iPSC signature gene expression differences are due to differential promoter binding by the reprogramming factors. High-resolution array profiling demonstrated that there is no common specific subkaryotypic alteration that is required for reprogramming and that reprogramming does not lead to genomic instability. Together, these data suggest that iPSCs should be considered a unique subtype of pluripotent cell.

A large number of mutations in the gene encoding the catalytic subunit of mitochondrial DNA polymerase Î³ (POLÎ³A) cause human disease. The Y955C mutation is common and leads to a dominant disease with progressive external ophthalmoplegia and other symptoms. The biochemical effect of the Y955C mutation has been extensively studied and it has been reported to lower enzyme processivity due to decreased capacity to utilize dNTPs. However, it is unclear why this biochemical defect leads to a dominant disease. Consistent with previous reports, we show here that the POLÎ³A:Y955C enzyme only synthesizes short DNA products at dNTP concentrations that are sufficient for proper function of wild-type POLÎ³A. In addition, we find that this phenotype is overcome by increasing the dNTP concentration, e.g. dATP. At low dATP concentrations, the POLÎ³A:Y955C enzyme stalls at dATP insertion sites and instead enters a polymerase/exonuclease idling mode. The POLÎ³A:Y955C enzyme will compete with wild-type POLÎ³A for primer utilization, and this will result in a heterogeneous population of short and long DNA replication products. In addition, there is a possibility that POLÎ³A:Y955C is recruited to nicks of mtDNA and there enters an idling mode preventing ligation. Our results provide a novel explanation for the dominant mtDNA replication phenotypes seen in patients harboring the Y955C mutation, including the existence of site-specific stalling. Our data may also explain why mutations that disturb dATP pools can be especially deleterious for mtDNA synthesis.

It has been assumed, based largely on morphologic evidence, that human pluripotent stem cells (hPSCs) contain underdeveloped, bioenergetically inactive mitochondria. In contrast, differentiated cells harbour a branched mitochondrial network with oxidative phosphorylation as the main energy source. A role for mitochondria in hPSC bioenergetics and in cell differentiation therefore remains uncertain. Here, we show that hPSCs have functional respiratory complexes that are able to consume O(2) at maximal capacity. Despite this, ATP generation in hPSCs is mainly by glycolysis and ATP is consumed by the F(1)F(0) ATP synthase to partially maintain hPSC mitochondrial membrane potential and cell viability. Uncoupling protein 2 (UCP2) plays a regulating role in hPSC energy metabolism by preventing mitochondrial glucose oxidation and facilitating glycolysis via a substrate shunting mechanism. With early differentiation, hPSC proliferation slows, energy metabolism decreases, and UCP2 is repressed, resulting in decreased glycolysis and maintained or increased mitochondrial glucose oxidation. Ectopic UCP2 expression perturbs this metabolic transition and impairs hPSC differentiation. Overall, hPSCs contain active mitochondria and require UCP2 repression for full differentiation potential.

UCP2 &#xC5D0;&#xB108;&#xC9C0; &#xB300;&#xC0AC; &#xBC0F; &#xBD84;&#xD654; &#xC870;&#xC808; &#xC778;&#xAC04;&#xC758; pluripotent &#xC904;&#xAE30; &#xC138;&#xD3EC;&#xC758; &#xC7A0;&#xC7AC;&#xB825;.

Se ha asumido, basado en gran parte en las pruebas morfol&#xF3;gicas, que las c&#xE9;lulas madre humanas pluripotentes (hPSCs) contienen mitocondrias subdesarrollado, bioenerg&#xE9;ticamente inactivo. En contraste, las c&#xE9;lulas diferenciadas puerto una red ramificada mitocondrial con la fosforilaci&#xF3;n oxidativa como la principal fuente de energ&#xED;a. El papel de las mitocondrias en la bioenerg&#xE9;tica hPSC y en la diferenciaci&#xF3;n celular por lo tanto, sigue siendo incierto. Aqu&#xED;, mostramos que tienen hPSCs funcionales complejos respiratorios que son capaces de consumir O (2) a m&#xE1;xima capacidad.
