在免疫荧光显微镜技术中,荧光标记的抗体在结合特定靶标或抗原后会发出荧光。

免疫荧光显微镜利用光激发荧光染料的电子进入更高的能级。当电子回到基态时,会释放出波长更长的光。

这种发射光或荧光可实现对组织内特定细胞或细胞内特定蛋白质的可视化观察。

当带有荧光标记的一抗直接结合靶蛋白时,称为直接免疫荧光。或者,也可采用间接免疫荧光,即带有荧光标记的二抗结合已与目标蛋白结合的一抗。间接法产生的荧光信号强于直接免疫荧光法。

直接免疫荧光法用于检测组织中异常蛋白质的聚集,而间接免疫荧光法则可用于在自身免疫性疾病诊断过程中检测血清中的循环抗体。
