摘要

引言

细胞间通信和信号传导对于组织和整个器官水平上响应细胞外激动剂的生理过程协调至关重要1,2。最直接的细胞间通信方式是由缝隙连接(gap junctions)的形成所介导的。缝隙连接是由多个缝隙连接通道组成的斑块结构,这些通道是由相邻细胞的两个连接蛋白(connexin, Cx)半通道以头对头方式对接形成的蛋白质通道3,4图1)。缝隙连接允许分子量小于1.5 kDa的小分子信号物质通过,包括Ca2+或IP35,从而引发并调节邻近细胞内钙库的Ca2+释放6图2)。缝隙连接通道受到细胞内和细胞间蛋白质相互作用以及细胞信号通路(如氧化还原修饰和磷酸化)的严密调控7。缝隙连接(GJs)促进相连细胞之间的协调反应,使其发挥化学和电同步合胞体的功能。例如,心房和心室肌细胞之间心脏动作电位的传播即由Cx构成的缝隙连接通道介导85。连接蛋白(Cxs)不仅作为缝隙连接通道发挥作用,还可形成未配对的半通道,在细胞膜上发挥类似常规离子通道的功能8-10图1)。半通道通过调控细胞内外环境之间的离子和信号分子交换,参与邻近细胞间的旁分泌信号传递。

方案

1. 角膜内皮细胞的分离

开始之前:从当地屠宰场获取的新鲜眼球中,在眼球摘除后尽快分离细胞。确保眼球来自一头死亡后5分钟内、年龄不超过18个月的牛,并在运输至实验室期间以4 °C的Earle's平衡盐溶液-1%碘溶液保存。

  1. 将眼球从Earle's平衡盐溶液-1%碘溶液中取出,放入培养皿(100 × 20 mm)中。
  2. 用含70%乙醇的溶液对眼球进行消毒,并用含1%碘的Earle's平衡盐溶液冲洗。
  3. 从本步骤开始,在无菌操作台中进行操作。小心地将角膜从眼球上分离,并将其放入含有Earle's平衡盐溶液的培养皿(35 × 10 mm)中,使上皮细胞层朝上。如有必要,小心去除仍附着在角膜上的残留虹膜组织。
  4. 将角膜转移至另一含有Earle's平衡盐溶液的培养皿中,使内皮细胞层朝上,并用Earle's平衡盐溶液冲洗两次。
  5. 将内皮层朝上的角膜转移至钟形杯(一种杯状培养皿)中,并加入生长培养基覆盖。生长培养基由含25 mM葡萄糖的Dulbecco改良Eagle培养基(DMEM)组成,其中包含10%胎牛血清、6.6% L-谷氨酰胺、2.5 μg/ml两性霉素B,以及含10,000单位/ml青霉素、10,000 μg/ml链霉素和25 μg/ml两性霉素B的1%抗生素-抗真菌混合液。
  6. 用吸液移液管吸除培养基。
  7. 向角膜的内皮层加入300 μl胰蛋白酶溶液(0.5 g/L)(所有涉及胰蛋白酶的步骤均使用相同浓度)。
  8. 将含有角膜的钟形杯放入加盖的培养皿中,置于37 °C、5% CO2的培养箱中孵育30分钟。
  9. 在无菌操作台中,用火焰抛光的钩状玻璃巴斯德吸管轻轻刮取角膜上的内皮细胞。
  10. 吸出含有内皮细胞的溶液,并将其加入含有4 ml培养基的培养瓶(25 cm2)中。
  11. 向角膜中加入300 μl生长培养基,重复刮取操作,并将含有内皮细胞的溶液加入培养瓶中。
  12. 将此最后一步(1.11)再重复一次。
  13. 将含有角膜内皮细胞并加入生长培养基的培养瓶置于37 °C、5% CO2的培养箱中培养。
  14. 两天后,加入6 ml培养基。
  15. 每两天更换一次生长培养基。

2. 细胞培养

  1. 当细胞融合度达到时(分离后约10天内),移除培养基,并用Earle's平衡盐溶液洗涤细胞两次。
  2. 向细胞中加入1.5 ml胰蛋白酶溶液以使细胞脱落,将培养瓶置于培养箱中(37 °C,5% CO2)孵育3至4分钟。
  3. 加入12 ml生长培养基。用移液器将液体反复吹打三次,以分散细胞并进行细胞计数。
  4. 根据细胞密度以不同比例接种细胞,并放入培养箱中。准备两个带孔的培养玻片(面积为4.2 cm2),每孔接种165,000个细胞(细胞密度为39,286个/cm2)。准备80 cm2的培养瓶用于新传代,接种密度为6,250个/cm2,并加入新鲜培养基至总体积为25 ml。
  5. 每两天更换一次培养基。
  6. 培养瓶中的细胞层在3至4天后达到融合状态,可用于实验。
  7. 当培养瓶中的细胞层达到融合状态时,重复步骤2.1至2.6。传代次数不超过2代的细胞可用于实验。

3. 诱导钙波的机械刺激

  1. 在37 °C、轻度振荡条件下,用含10 μM Fluo-4 AM的磷酸盐缓冲液将细胞加载至腔室载玻片中,孵育30 min。
  2. 移除Fluo-4 AM溶液,用磷酸盐缓冲液洗涤细胞5次,随后将细胞在磷酸盐缓冲液中孵育,并在室温下静置至少5 min后再进行测量。
  3. 使用氩离子激光器在488 nm处激发,采用HFT 488分光镜,通过长通发射滤光片LP 505在530 nm处收集荧光发射信号,针孔设为最小。使用40X油浸物镜(空气,1.2 N.A.)。在ARL-67156实验中,使用10X物镜(空气,0.3 N.A.)。
  4. 在共聚焦显微镜下寻找细胞达到完全融合的视野。
  5. 将移液管定位至与腔室载玻片成45°角,并接触细胞膜。对单个细胞施加短暂(≈1秒)的机械刺激。该机械刺激是通过连接在压电晶体纳米定位器上的玻璃微移液管(尖端直径<1 μm)短暂接触小于1%的细胞膜,引起急性且持续时间短的细胞形变,纳米定位器通过安装在显微操作器上的放大器控制。玻璃微移液管由微电极拉制仪制备。确保在机械刺激过程中,纳米定位器的工作电压保持在0.2至1.5 V之间。电压超过1.5 V可能导致细胞损伤。对于每种细胞类型和实验条件,必须通过从低(0.2 V)到高(1.5 V)的一系列电压测试,仔细确定在不损伤细胞的前提下进行机械刺激的最佳电压。电压是刺激力度的度量指标,因为它决定了施加在细胞膜上的机械应力和应变。机械刺激的力可通过机械应力乘以面积计算得出。由于面积(<3.14 μm2)和机械应力均非常小,因此机械刺激的力也很小。(注意:当细胞受损时,荧光会从细胞中泄漏,细胞变暗。)
  6. 使用共聚焦显微镜测量机械刺激后[Ca2+]i的空间变化。
  7. 采集并存储图像。
  8. 利用共聚焦显微镜的软件绘制多边形感兴趣区域,以定义响应细胞的总表面积(活性区域,AA)。
