摘要

跨分类界域的多微生物相互作用研究,迄今尚未探讨微生物组中的病毒成分如何影响其他微生物与病毒感染宿主细胞之间的后续相互作用。病毒与细菌及真菌的共存主要存在于口腔和生殖道的黏膜表面。黏膜细胞,特别是那些存在持续性慢性或潜伏性病毒感染的细胞,可能通过病毒介导的受体表达数量和类型改变,显著影响微生物组成员。宿主细胞膜结构的改变将导致正常菌群及机会性病原体在生物膜形成初始阶段的附着能力发生变化。,黏附。本研究描述了一种定量检测和直观观察HSV对生物膜形成初期(黏附)影响的方法 S. aureusC. albicans.

引言

人类微生物组包含来自多个分类界别的多种生物,它们在人体内共享特定的地理区域。附着于细胞表面是生物膜形成的关键第一步,而生物膜形成是微生物组定植过程的一部分。微生物组中可能还包括引起慢性和持续性感染的病毒。这些病毒对细胞的慢性感染可能导致假定受体可用性的改变1,2。此外,胞内病原体进入细胞也可能影响宿主细胞膜的流动性/疏水性,从而进一步改变其他微生物组成员(包括细菌和真菌)的附着能力。为了理解这些共定位于人体相同区域的多种病原体之间可能发生的相互作用,我们必须能够研究代表黏膜表面存在的各个分类界病原体之间的相互作用。

方案

1. 单纯疱疹病毒(HSV)毒株及其处理

注意:所使用的带有β-半乳糖苷酶报告基因活性的重组非扩散型HSV-1(KOS) gL86和HSV-2(KOS) 333gJ-由V. Twiari提供36,37

  1. 使用同一批次的病毒,以1:1的比例与含20%胎牛血清(FBS)和脱脂奶粉的杜氏改良伊格尔培养基(DMEM)混合,于-80 °C保存备用。在病毒批次储存前,通过以下方法测定病毒浓度: o邻硝基苯基-β-D-半乳糖苷(ONPG)和5-溴-4-氯-3-吲哚基-β-D-半乳糖苷(X-Gal)检测
  2. 通过报告病毒进入实验,使用X-Gal染色法测定每次实验中病毒的感染活力及感染复数(MOI),具体步骤如前所述(图1)14.
  3. 将病毒用Opt-MEM稀释至所需MOI。染色前用多聚甲醛(0.5 ml/孔)固定单层细胞。将病毒活性对照与多微生物检测板平行设置于另一块微量培养板中。

2. HeLa 299 细胞操作

  1. 在含4.5 g/L葡萄糖、10%灭活胎牛血清(FBS)、庆大霉素(50 µg/ml)和L-谷氨酰胺的DMEM培养基中,于37 °C、5% CO2条件下培养。当细胞融合度达到80%时,使用胰蛋白酶溶液(乙二胺四乙酸,0.53 M EDTA;0.05%胰蛋白酶;5 ml/瓶)进行传代。

3. C. albicans 操作处理

注意:C. albicans 来自临床实验室来源,保存于-80 °C的Remmel脱脂牛奶2x培养基中。

  1. 将冻存菌种接种于沙氏葡萄糖培养基中(37 °C)培养。24 小时后,将白色念珠菌(C. albicans)转接至Fungisel培养基(37 °C,48 小时),用于后续实验。
  2. 诱导生成芽管(GT)形态(挑取典型菌落,加入3 ml胎牛血清,37 °C孵育3小时,OD600值为0.3)。孵育结束后,用HBSS缓冲液洗涤芽管细胞两次(4,000 × g离心)。将洗涤后的芽管重悬于预热的HBSS缓冲液中(37 °C,OD600值为0.32)。经血细胞计数板镜检确认,芽管形态应占观察细胞总数的99%。
  3. 制备酵母形态(YF)菌悬液:挑取典型Fungisel培养基上的菌落,重悬于HBSS缓冲液中(3 ml,OD600值为0.32)。使用血细胞计数板在显微镜下计数每毫升中的酵母形态细胞数量。经血细胞计数板镜检确认,酵母形态应占观察细胞总数的99%。
  4. 制备工作用真菌菌液(取250 µl芽管或酵母形态菌液加入25 ml HBSS缓冲液中,37 °C孵育,终浓度为105 CFU/ml)。

4. S. aureus 操作处理

  1. S. aureus ATCC 25923 储存于-80 °C(脱脂牛奶保护剂,2倍浓度)。在绵羊血琼脂培养基(5%;37 °C;24 小时)上进行培养。挑选典型菌落,并于2天内转移至甘露醇盐培养基中制备菌种(37 °C;18 小时)。
  2. 制备 S. aureus 菌液储备液(3 ml HBSS;光密度Abs600为1.32,浓度为108 CFU/ml)
    1. 制备工作用 S. aureus 储备菌液(取100 µl 储备菌液加入25 ml HBSS中,终浓度为105 CFU/ml)。

5. CandidaS. aureus 悬液

  1. 制备混合的 C. albicansS. aureus 悬液(将250 µl YF或GT储备液与100 µl S. aureus 储备液加入25 ml HBSS中)。

6. 多微生物生物膜检测

  1. 在96孔板中接种200 µl浓度为2 × 105 个HeLa细胞/ml的细胞悬液(汇合度达85%)。轻轻摇动培养板(30–45分钟;37 °C),然后置于培养箱中孵育(37 °C;5% CO2,18小时)。用Opt-MEM洗涤单层细胞一次,随后在每孔中加入100 µl Opt-MEM稀释的HSV病毒(HSV-1 (KOS) gL86 或 HSV-2 (KOS) 33 gJ-),感染复数(MOI)分别为50和10。将培养板置于37 °C、5% CO2条件下孵育3小时。每天仅使用一种病毒株。
  2. 用含Mg+2和Ca+2的磷酸盐缓冲液(PBS)洗涤感染后的单层细胞一次(100 µl),弃去PBS后每孔保留25 µl,再加入预温的HBSS。
    1. 根据表1所示,加入YF、GT和/或S. aureus的工作悬液(100 µl;靶细胞与细胞的比例为5:1;n=16)。将培养板静置孵育(30分钟;37 °C;5% CO2)。
  3. 孵育结束后,逐列吸去液体,并立即用300 µl含Mg+2和Ca+2的PBS重新填充。重复此洗涤步骤两次,然后每孔加入200 µl经滤膜除菌的放射性免疫沉淀分析裂解缓冲液(RIPA;1:50稀释)。
  4. 迅速吹打裂解液以充分混匀,取50 µl滴加至甘露醇盐琼脂(MS)和/或沙氏葡萄糖琼脂(Fungisel,F)培养基上(参见图2)。使用90°弯角的玻璃涂布棒将裂解液均匀涂布。将培养基置于37 °C孵育18小时后,人工计数每块平板上的菌落数。对照组包括S. aureus和/或C. albicans在未感染HSV的HeLa细胞上的黏附情况。

7. 影像学研究

  1. 用50 ml丙酮(置于100 ml烧杯中)清洗每片圆形玻璃盖玻片(12 mm)。用Kimwipes纸擦干并灭菌盖玻片(置于玻璃培养皿中)。用经酒精火焰灭菌的镊子将干燥且无菌的盖玻片放入无菌24孔板的各孔中。
  2. 将HeLa细胞(1 ml;96孔板所用体积的5倍)加入含有经洗涤的无菌圆形玻璃盖玻片的24孔板各孔中。根据模板加入病毒、细菌和真菌。 (表2) 使用96孔板所需体积的5倍量,然后按照上述步骤6.2.1至6.4所述进行孵育和处理。
  3. 最后一次洗涤后,用甲醇浸没载玻片以固定细胞,并让甲醇自然挥发。将载玻片于室温保存,直至染色。
  4. 对于明场显微镜观察(最终放大倍数1,000倍),向含有甲醇固定的盖玻片的孔中加入去离子水。立即吸去水分。每张盖玻片上滴加革兰氏结晶紫染液。用去离子水洗涤盖玻片,去除未结合的染料。干燥盖玻片 原位 然后使用硬封固介质将它们粘附到已标记的载玻片上 (图3).
  5. 对于荧光显微镜观察(100倍物镜;最终原始放大倍数1,000倍),按照步骤7.4所述基本方法用缓冲液洗涤盖玻片以去除甲醇。将盖玻片在孔板中干燥。
    1. 干燥后,取下盖玻片并将其置于标记好的载玻片上。然后加入足量的1:20稀释的异硫氰酸荧光素(FITC)标记的单纯疱疹病毒1型和2型gD抗体溶液,以覆盖盖玻片(1–5 µl)。
    2. 在湿盒中于 37 °C 孵育载玻片 30 分钟。孵育结束后,用 PBS 洗涤盖玻片,更换 4 次。
    3. 最后一次洗涤后,将盖玻片放回已标记的载玻片上。用4′,6-二脒基-2-苯基吲哚(DAPI;湿盒中,37 °C孵育30分钟)染色。孵育结束后,用PBS更换4次洗涤盖玻片,然后用硬封固封片剂将其固定于已标记的载玻片上。
    4. 让封片剂在室温避光条件下固化24小时。使用亮场显微镜或配备FITC和DAPI截止滤光片的荧光显微镜,通过油镜观察盖玻片。至少观察50个视野(每种生物不少于100个细胞)的图像,进行共定位分析。图4).

结果

本报告所述系统所获得数据的稳健性水平见图2 a-f 38。利用该系统,可阐明葡萄球菌与真菌同病毒感染细胞之间的相互作用及其对彼此黏附能力的影响。此类研究需要进行显微镜检查,如图3图438所示,以确定多微生物相互作用是否发生在同一细胞上。本研究中观察到的差异性细胞相互作用,是HSV对葡萄球菌和真菌黏附能力进行调节的结果,且该调节具有病毒种类特异性。
