通过口服递送双链RNA在韧皮部及刺吸式口器昆虫中诱导RNA干扰的方法

本文展示了通过植物维管组织口服递送双链RNA(dsRNA)以实现韧皮部汁液取食昆虫RNA干扰(RNAi)的新型技术。

摘要

韧皮部及植物汁液取食性昆虫通过侵入农作物和果实以获取营养,在此过程中对粮食作物造成损害。半翅目昆虫是一类重要的农业害虫,通过取食韧皮部汁液对作物造成破坏。其中,褐臭蝽(BMSB), Halyomorpha halys (异翅亚目:蝽科)和亚洲柑橘木虱(ACP), Diaphorina citri Kuwayama(半翅目:Liviidae)是一类被引入北美的半翅目昆虫害虫,在当地已成为多种高价值经济作物、大田作物、主粮作物以及柑橘类水果的重要入侵性农业害虫;当其在室内聚集时,亦成为令人困扰的滋扰性害虫。由于多种害虫已对杀虫剂产生抗性,促使人们开发替代性的害虫管理策略。双链RNA(dsRNA)介导的RNA干扰(RNAi)是一种用于功能基因组学研究的基因沉默机制,同时具备作为害虫管理工具的应用潜力。外源合成的双链RNA或小干扰RNA(siRNA)可通过降解与之序列同源的内源RNA,引发高效特异的基因沉默。要将RNAi有效且环境友好地应用于半翅目昆虫的生物防治,作为分子生物杀虫剂使用,需要 体内 通过饲喂方式递送dsRNA。本文展示了将dsRNA递送至昆虫的方法:通过浸渍法将dsRNA载入青豆,以及通过口服摄入吸收基因特异性dsRNA。我们还概述了非转基因植物递送策略,包括叶面喷施、根部浇灌、树干注射以及黏土颗粒载体,这些方法均有助于dsRNA的持续释放。经口服摄入递送dsRNA被证实可有效达到剂量要求,显著降低靶基因的表达水平,例如保幼激素酸O-甲基转移酶(JHAMT)和卵黄原蛋白(Vg)。这些创新性方法为利用dsRNA进行作物保护及应对害虫治理中的环境挑战提供了可行策略。 关键词:dsRNA递送,饲喂法,青豆浸渍,口服摄入,叶面喷施,根部浇灌,树干注射,黏土颗粒,非转基因植物,RNA干扰,作物保护,害虫管理

引言

半翅目昆虫因其能够迅速大量繁殖并在植物间传播病害,已成为对农业具有重大经济影响的害虫。褐臭虫(H. halys Stål)是一种入侵性害虫,于1996年首次被报道在宾夕法尼亚州阿伦敦发现,其原产于亚洲(中国、中国台湾、韩国和日本),是意外引入西半球的1。自引入以来,褐臭虫已在43个州被发现,其中以中大西洋地区(特拉华州、马里兰州、宾夕法尼亚州、新泽西州、弗吉尼亚州和西弗吉尼亚州)种群密度最高,同时也在加拿大和欧洲出现,对农业生产构成潜在威胁2。作为一种多食性害虫,褐臭虫可危害约300种已知的寄主植物,包括苹果、葡萄、观赏植物、种子作物、大豆和玉米等高价值作物。其危害主要源于一种称为“刺破并冲洗”的取食方式,即利用针状口器刺入寄主作物,获取维管组织中的营养物质2,3。此外,褐臭虫也是一种室内害虫,在秋季至冬季期间可能进入学校和住宅等生活区域栖息2。研究表明,褐臭虫释放的化学物质和空气传播的过敏原可引发果树作物作业人员的过敏反应,还可能导致敏感个体出现接触性皮炎、结膜炎和鼻炎等过敏性疾病4,5。另一种半翅目昆虫——柑橘木虱(D. citri Kuwayama,半翅目:Liviidae),是柑橘类水果的重要害虫,可传播局限于韧皮部的细菌(Candidatus Liberibacter asiaticus),引发黄龙病(HLB),即广为人知的柑橘黄化病6,7。黄龙病最早在中国南部被报道,目前已蔓延至亚洲、非洲、大洋洲、南美洲和北美洲共40个不同国家7。柑橘黄化病是全球性难题,因柑橘果实大量损失而造成严重的经济和财政损失,因此,控制柑橘木虱被视为预防和治理黄龙病的重中之重。

有效控制这些害虫的措施通常需要使用持效期相对较短的化学杀虫剂。化学杀虫剂防治策略往往缺乏安全的环境管理措施,或由于害虫种群对杀虫剂产生抗性而导致防治效果下降8,9。因此,利用分子生物农药进行害虫生物防治是一种潜在的替代方案,但其在全球范围内的应用仍较为有限,而多种寄生性天敌(例如Trisolcus japonicus)也可能作为有效的天然生物防治手段。RNA干扰(RNAi)是一种具有潜力的新兴技术,可用于利用分子生物农药防控入侵性害虫10。RNAi是一种已被充分阐明的基因调控机制,能够以序列特异性方式有效实现内源性以及外源双链RNA(dsRNA)的转录后基因沉默,最终在mRNA水平上调控基因表达11,12。简而言之,当外源dsRNA进入细胞后,由RNase III超家族中一种称为Dicer的双链特异性核酸酶将其加工成小干扰RNA(siRNA),Dicer蛋白在进化上保守存在于线虫、果蝇、植物、真菌和哺乳动物中13,14,15。这些长度为21–25个核苷酸的siRNA双链随后解旋,并作为引导RNA整合到RNA诱导的沉默复合体(RISC)中。该RISC-RNA复合物可通过Watson-Crick碱基配对识别互补的靶标mRNA,最终由含有RNase H样结构域的多结构域蛋白Argonaute介导mRNA的切割,导致相应mRNA降解并减少蛋白质翻译,从而实现转录后基因沉默16,17,18

利用RNAi进行害虫管理需要引入dsRNA 体内 通过沉默目标基因,从而激活siRNA通路。已用于向昆虫及昆虫细胞递送dsRNA以诱导系统性RNAi的方法包括饲喂法10,19,浸泡20,21,显微注射22,如脂质体等载体23以及其他技术24RNAi 最早是在 秀丽隐杆线虫 沉默 unc-22 Fire 和 Mello 的基因表达研究25,随后敲低frizzled基因的表达 黑腹果蝇26最初的 功能 研究 采用 显微注射 技术 将 dsRNA 递送至 昆虫 体内,例如 Apis mellifera22,27, Acyrthosiphon pisum28, 德国小蠊29, H. halys30和鳞翅目昆虫(由Terenius综述) 31)。显微注射的优势在于能够向昆虫目标部位递送准确且精确的剂量。尽管此类有菌穿刺可能因创伤而诱导免疫相关基因的表达32因此,排除了其在农业生物农药开发中的实用性。

另一种递送dsRNA的方法 体内 是通过浸泡法实现的,该方法涉及将动物或细胞悬浮于含有dsRNA的细胞外培养基中,使其摄取或吸收dsRNA。浸泡法已被有效用于诱导RNAi 果蝇 S2 细胞培养以抑制 Raf1 下游(Downstream-of-Raf1)DSOR1丝裂原活化蛋白激酶激酶(MAPKK)20以及在 秀丽隐杆线虫 以沉默 pos-1 基因33然而,与显微注射相比,通过浸泡法递送的dsRNA在诱导RNAi方面的效率较低。20. 首次在咀嚼式口器昆虫中实现RNAi介导的基因沉默是在西方玉米根虫(WCR)中证实的(Diabrotica virgifera virgifera通过将dsRNA注入人工琼脂饲料中10早期的报告已总结了将dsRNA注入节肢动物特异性天然饮食中的递送方法34这些递送方式被进一步证实与人工递送方法具有相当的效率,例如采采蝇(tsetse fly)的情况Glossina morsitans morsitans),在通过血液餐或显微注射递送dsRNA时,均观察到免疫相关基因的等效敲低35同样,通过液滴递送dsRNA至褐卷蛾(light brown apple moth)Epiphyas postvittana)36,小菜蛾(Plutella xylostella幼虫37,以及蜜蜂38,39 诱导高效的RNAi。在半翅目昆虫中,最有效的RNAi实验大多采用注射dsRNA的方法40 因为半翅目昆虫的dsRNA口服递送较为困难,必须通过寄主植物的维管组织进行传递。在亚洲柑橘木虱(ACP)和玻璃翅叶蝉(GWSS)中也观察到了有效的RNA干扰现象。 Homalodisca vitripennis通过根部浇灌、叶面喷施、树干注射或插条吸收,将dsRNA递送至柑橘和葡萄植株的维管组织中41,42,43,44,45,46这也促成了针对ACP的dsRNA的首个专利(2016年,美国专利号US 20170211082 A1)。利用纳米颗粒和脂质体等载体递送siRNA和dsRNA可提高其稳定性,且增强dsRNA递送效率的研究进展迅速。23,47,48,49,50一种新型基于纳米颗粒的核酸递送载体 体外 体内 专门针对治疗应用而总结的可能作为合适的递送载体具有巨大潜力51纳米颗粒作为dsRNA的递送载体可能存在一些缺点,包括溶解性差、疏水性强或生物累积能力有限52,但合适的递送辅助聚合物可弥补这些缺点。自递送核苷酸的开发与应用也正在兴起,这类物质称为“反义寡核苷酸”,为单链RNA/DNA双链体46.

节肢动物中的卵黄发生是一个调控繁殖的关键过程,受保幼激素(JH)或蜕皮素的调节,这两种激素是脂肪体合成卵黄原蛋白(Vg)的主要诱导因子;Vg最终通过Vg受体介导的内吞作用被发育中的卵母细胞摄取53。Vg是一类在卵巢外合成的多肽,对主要卵黄蛋白——卵黄蛋白(vitellin)的形成至关重要54,55,因此在繁殖和衰老过程中具有重要作用56。Vg已在线虫57以及蜜蜂(Apis mellifera)中成功实现沉默,在后者中,通过RNAi介导的Vg耗竭在成虫和卵中均被观察到22。由于推测Vg的耗竭将导致可观察的表型效应,如生育力和繁殖力下降,从而可能有助于对BMSB的防控,因此开展了RNAi介导的Vg转录后基因沉默实验。JHAMT基因编码S-腺苷-L-甲硫氨酸(SAM)依赖性的JH酸O-甲基转移酶,催化JH生物合成途径的最后一步58。在该途径中,法尼基焦磷酸(FPP)依次由法尼醇转化为法尼酸,再经JHAMT将甲基法尼酸酯转化为JH。该途径在昆虫和节肢动物中高度保守,特异性参与变态发育过程,而这一过程由激素在发育上进行调控59,60,61。在B. mori中,JHAMT基因的表达及咽侧体中的JH生物合成活性表明,JHAMT基因的转录抑制对终止JH的生物合成至关重要58。因此,选择JHAMTVg基因作为RNAi靶向耗竭的目标。此外,RNAi也被用于柑橘树中以控制ACP和GWSS。通过根部浇灌、茎干滴注(树干注射)以及叶面喷施等方式,向柑橘树施加针对昆虫特异性精氨酸激酶(AK)转录本的dsRNA42,44。dsRNA的外用处理在柑橘树冠各部位均被检测到,表明其通过植物维管组织实现了有效递送,并导致ACP和GWSS的死亡率显著升高41,42,45

在本研究中,我们确定了一种用于递送dsRNA等治疗物质的天然饮食途径。随后,利用基因特异性的dsRNA,通过该新开发的技术在褐蝽若虫中实现了JHAMT和Vg mRNA的沉默,如先前研究所示62。本文所展示的这些新型递送方案优于传统的RNA递送系统,后者通常依赖于表面喷雾或显微注射。蔬菜和水果、茎部渗出液、土壤浇灌以及黏土吸附剂均可用于dsRNA的递送,这对于生物农药在害虫与病原体管理中的持续发展至关重要。

方案

1. 棒蝽饲养

  1. 按照标准实验室操作和先前所述方法饲养BMSB昆虫63
  2. 在温室(22 °C)和自然光照条件下,以Citrus macrophylla饲养ACP(D. citri)昆虫。使用羽化后约5–7天的成虫ACP。

2. 基因区域的选择与 体外 dsRNA的合成

  1. 从先前发表的转录组谱中筛选出BMSB特异性基因32.
  2. 确保所选目标区域在200至500个碱基对之间。
  3. 使用下述条件对基因组DNA进行聚合酶链式反应(PCR),以扩增目标基因的相关片段。参见 表1 用于基因特异性寡核苷酸。
    1. PCR反应:在0.25 mL的PCR管中,加入5 µL 10X PCR缓冲液、4 µL dNTP混合液(各2.5 mM)、2 µL DNA模板(50 ng/µL)、引物1和引物2各2.5 µL(10 µM)、0.25 µL DNA聚合酶(5 U/µL),并用无DNase/RNase的水补足至50 µL。
    2. PCR条件:将PCR反应在95 °C下预变性3 min,随后进行30个循环,每个循环包括98 °C变性10 s、55 °C退火30 s、72 °C延伸1 min;最后在72 °C继续延伸10 min。使用纯化试剂盒对PCR产物进行纯化。
  4. 使用基因特异性引物进一步扩增获得的PCR片段,引物两端需包含T7 RNA聚合酶启动子序列(5'-GAA TTA ATA CGA CTC ACT ATA GGG AGA-3'),具体操作如前所述62.
  5. 使用 LacZ 基因作为 RNAi 的阴性对照(模拟对照)。
    注意:LacZ 是一个编码 β-半乳糖苷酶的基因,该酶由 大肠杆菌 基因组DNA(所用引物列于 表1).
  6. 执行 体外 转录以生成dsRNA,方法如前所述62.
  7. 将所得的dsRNA溶解并重悬于150 µL无DNase/RNase的水中,测定浓度后于-80 °C保存,以备后续使用。

3. 利用青豆递送dsRNA

  1. 选择早期4th 来自同一卵块的二龄BMSB若虫,在进行dsRNA饲喂前饥饿处理24小时。
  2. 选择细长的经认证的有机青豆(Phaseolus vulgaris L.),用0.2%次氯酸钠溶液清洗5分钟。
    注意:选择细长的青豆,以便青豆能轻松放入2 mL 微型离心管中。
  3. 用ddH₂O洗涤3次2O,并静置晾干。
  4. 使用洁净的剃须刀片从豆荚末端切除,使四季豆总长度为7.5 cm。
  5. 将清洗并修剪好的青豆浸入装有300 µL对照溶液(绿色食用色素按1:10稀释,成分:水、丙二醇、Fd&C 黄色 5,Fd&C 蓝 1,以及对羟基苯甲酸丙酯作为防腐剂)。
  6. 进行稀释的步骤 体外 通过将5 µg或20 µg的LacZ、JHAMT或Vg dsRNA溶解于300 µL无RNase/DNase的水中,分别配制成终浓度为0.017 µg/µL或0.067 µg/µL的溶液。
  7. 将清洗并修剪好的青豆浸入装有300 µL dsRNA溶液(来自步骤3.6)的无盖2 mL微量离心管中。
  8. 用封口膜包裹并密封装有浸泡豆子的微量离心管管口,以防止dsRNA溶液蒸发,并阻止昆虫进入微量离心管内。
  9. 将这些试管在室温下直立放置3小时,使双链RNA溶液通过毛细作用充分渗透至整个绿豆中。
  10. 将这些试管放入洁净的培养容器(聚丙烯材质)中。放入三只饥饿处理的4th 培养容器中的若虫龄期BMSB若虫。
  11. 每个培养容器中放入三只昆虫,并加入三根含绿色食用色素或dsRNA溶液的青豆,每容器处理三只昆虫。将昆虫置于培养箱中,保持25 °C、72%相对湿度,光照周期为16小时光照:8小时黑暗。
  12. 让昆虫取食浸泡并吸收了dsRNA的青豆5天,但在3天后需更换为经dsRNA处理的新鲜青豆。

4. 基于RNAi介导的基因沉默后,利用实时定量(qPCR)分析BMSB中的基因表达

  1. 通过qPCR检测RNAi对转录本表达水平的影响。
  2. 提取经dsRNA处理动物的总RNA,并合成cDNA62
  3. 使用实时PCR系统和表1中列出的引物进行qPCR反应。qPCR扩增条件如下:95 °C预变性10 min,随后进行40个循环(95 °C变性15 s,60 °C退火1 min),最后进行溶解曲线分析(95 °C 15 s,60 °C 1 min,95 °C 15 s,60 °C 15 s)。
  4. 确定qPCR标准曲线:以从正常动物总RNA制备的cDNA系列稀释液作为定量参考标准。
  5. 使用BMSB 18s RNA作为内参标准,校正不同组织间RNA回收量的差异32

5. 大型盆栽柑橘树和幼苗的叶面喷施应用

注意:柑橘品种“Carrizo”枳橙(Citrus sinensis × Poncirus trifoliata,芸香科)植株在玻璃温室中于自然光照和温度条件下培养,种植于1.2 L的容器中。植株持续进行修剪,以促进新的叶片嫩梢(称为“抽梢”)生长。柑橘木虱(ACP)倾向于在柑橘的新梢上取食和产卵64

  1. 选择植株或幼苗,并在使用前2-3天停止浇水,使土壤干燥至微湿但不完全干透的状态。
  2. 使用手动喷雾瓶将200 mL的dsRNA溶液(0.5 mg/mL)喷施到植株下部冠层(图4A)。
    注意:上述dsRNA溶液需用无DNase/RNase的水配制。
  3. 喷施后,让dsRNA溶液被叶片完全吸收。
    注意:柑橘树吸收了施用的dsRNA,随后采集施用前已被覆盖的新梢或枝条上的叶片;通过qPCR可检测到dsRNA,并显示其在3-4小时内系统性地迁移至树冠顶部叶片44,45,46
  4. 在修剪顶部处理25-40天后,从先前修剪的植株上采集新梢样本,取四个枝条顶端约10片叶子。提取总RNA,并利用表1所列引物及先前描述的方法46,通过反转录PCR(RT-PCR)和qPCR分析施用的dsRNA触发子的存在情况。
    注意:样品采集、总RNA提取、RT-PCR和qPCR操作均按先前所述方法进行46
  5. 类似地,将10 mL dsRNA溶液局部喷洒于幼苗或小型盆栽植株的下部叶片区域。
    注意:半翅目昆虫(ACP和GWSS)通常在处理后24小时获得取食机会,取食对象为未直接受喷施的新梢(叶片)、数周后新生的叶片,或整株植物。取食后第3、6和10天,检测发现昆虫体内呈dsRNA阳性。

6. 大型或小型盆栽柑橘树及幼苗的土壤/根部浇灌施用方法

  1. 选择植株或幼苗,并在使用前 2-3 天停止浇水,使土壤干燥至微湿但不完全干透的状态(这有助于形成可容纳待施加液体的空间)。
  2. 向大型盆栽植株(约 2.5 m 高)的土壤中加入 1 L 的 dsRNA 溶液(0.2 mg/mL),1 小时后再加入 1 L 水(作为冲洗液)。
  3. 向 1 m 高的盆栽树木的微干土壤中施加 100 mL 的 dsRNA 溶液(1.33 mg/mL)。
  4. 对于小型幼苗,向锥形容器中的土壤或裸露根部施加 10 mL 的 dsRNA 溶液(1 mg/mL)(图 4B C)。
  5. 让接受土壤灌注 dsRNA 溶液的植株浸泡 30 分钟,然后仅施加清水以促进根部吸收(黄色容器中的植株施加 20 mL,若较大的花盆容量 > 1 加仑,则施加 100 mL)。
    注意:将 dsRNA 局部施用于叶片后,可在处理后 3–6 小时内在枝条最远端检测到其存在,表明 dsRNA 在树木体内具有系统性移动能力。处理后 60–90 天的新枝检测结果仍呈 dsRNA 阳性。将枝条切段用于 dsRNA 饲喂生物测定实验,以提供给昆虫(ACP 和 GWSS)44

7. 大型盆栽柑橘树及幼苗的茎干注射(树干输液)施用方法

  1. 选择柑橘幼苗、新植株或约3.5年生的植株,用于通过茎部打孔(树干注射)方法注入dsRNA。
  2. 使用电钻和10 mm钻头在柑橘植株上钻孔,注意深度不要超过2 cm,或约为茎干直径的一半。
  3. 用宽0.6 cm(¼英寸)的密封膜条将每个注射器的铜尖端缠绕4–6圈,以防止尖端附近发生泄漏。
  4. 将6 mL(1.7 mg/mL)溶于无DNase/RNase水中的dsRNA溶液(此处标记为有色溶液)注入树干注射器中。
  5. 将溶液注入树干,并将注射器保留在树干内6–10小时,以确保dsRNA溶液被充分吸收。分别在处理后第3、10和30天,让昆虫取食经处理植株的枝条。
    注:通过树干注射法注入的dsRNA在植株体内可持续存在30–60天41,42,44。RNA干扰效果的验证采用qPCR进行,所用引物列于表1中。

8. 通过土壤递送至昆虫的经dsRNA处理的黏土颗粒

  1. 将黏土吸附剂倒入50 mL锥形管中,至管壁35 mL刻度线处(约30 g黏土吸附剂)。
  2. 向管中加入20 mL用无DNase/RNase水稀释的dsRNA溶液(100 µg/mL),以润湿全部吸附剂。盖紧锥形管盖子,轻轻倾斜管体以帮助排出空气,然后重新盖紧管盖。
  3. 将管子垂直放置,静置1–2分钟,使黏土颗粒充分吸收溶液。
  4. 将足量已吸附dsRNA的黏土加入土壤混合物中,填满一个1加仑的花盆。
  5. 用手充分搅拌并翻动土壤,使吸附dsRNA的黏土均匀分散于土壤中。
  6. 使用该土壤对选定用于dsRNA处理的幼苗进行移栽。
  7. 用200 mL不含dsRNA的清水浇灌土壤。30分钟至1小时后,再补充100 mL清水。24小时后,将植株转入常规浇水程序。
  8. 处理后每月采集经处理盆栽植株最新长出的顶端叶片4–6片,利用黏土吸附剂和dsRNA检测叶片中dsRNA的存在。
    注意:处理后24小时以上的植株或插条即可饲喂昆虫,且已证实可向昆虫持续递送RNA干扰效应长达一年(未发表数据)。

结果

RNAi 已被证明是研究基因生物学功能和调控的重要工具,在害虫管理方面具有巨大应用潜力19,68,69,70,71。针对特定昆虫物种,选择合适的靶基因进行沉默,以及确定相应的dsRNA递送方法,均至关重要。将dsRNA递送至昆虫体内的最佳方法必须通过实验确定,同时需优化递送剂量,因为不同方法可能各自具有优势和局限性。为了开发出最终可用于环境释放的分子生物杀虫剂,必须具备一种可行、高效且具有优势的递送方式。例如,外敷施用dsRNA已被证明在柑橘和葡萄上的昆虫防控中具有良好的递送效果42,44。一些创新策略,如利用饵剂、蔗糖溶液、酵母或细菌生产dsRNA供昆虫直接摄食,将dsRNA外敷喷洒于植物表面,或...
