一种利用嵌入金纳米颗粒的膜过滤器选择性去除放射性碘阴离子的高效方法

本文描述了一种利用固定化金纳米颗粒的醋酸纤维素膜滤器,对多种水溶液中的放射性碘进行快速、离子选择性脱盐的高效方法。

摘要

本文展示了一种用于制备嵌入纳米材料的复合膜及其在高效、离子选择性去除放射性碘中的应用的详细方案。利用柠檬酸盐稳定的金纳米颗粒(平均直径:13 nm)与醋酸纤维素膜,可简便地制备出金纳米颗粒嵌入的醋酸纤维素膜(Au-CAM)。Au-CAM上的纳米吸附剂在高浓度无机盐和有机分子存在下表现出高度稳定性。该工程膜可快速吸附水溶液中的碘离子。通过使用含Au-CAM的过滤装置进行过滤,可在短时间内实现优异的去除效率(>99%)以及离子选择性脱盐效果。此外,Au-CAM在不显著降低性能的情况下表现出良好的可重复使用性。这些结果表明,采用该工程化杂化膜的技术有望成为从液体废物中大规模去污放射性碘的有效方法。

引言

几十年来,医疗机构、科研设施和核反应堆产生了大量放射性液体废物。这些污染物常常对环境和人类健康构成切实威胁1,2,3。尤其是放射性碘,被公认为核电厂事故中最危险的元素之一。例如,关于福岛和切尔诺贝利核反应堆的环境报告表明,释放到环境中的放射性碘(包括131I(t1/2 = 8.02 天)和129I(t1/2 = 1570万年))的量超过了其他放射性核素4,5。特别地,这些放射性同位素的暴露会导致在人体甲状腺中高度摄取和富集6。此外,由于放射性碘在水中具有高溶解性,其释放可导致土壤、海水和地下水的严重污染。因此,已开展了大量研究,采用各种无机和有机吸附剂来捕获含水废物中的放射性碘7,8,9,10,11,12,13,14,15,16,17,18,19,20。尽管在先进吸附系统开发方面已投入大量努力,但在连续流动条件下具有满意性能的去污方法仍十分有限。最近,我们报道了一种新型脱盐工艺,该工艺利用由金纳米颗粒(AuNPs)构成的杂化纳米复合材料,表现出良好的去除效率、离子选择性、可持续性和可重复使用性21,22,23。其中,嵌入金纳米颗粒的醋酸纤维素膜(Au-CAM)在连续流动系统下对碘离子的脱盐效率显著高于现有吸附材料。此外,整个过程可在短时间内完成,这对于处理医疗和工业应用后产生的核废料而言是另一优势。本文的总体目标是提供Au-CAM制备的逐步操作方案24。我们还展示了一种利用工程化复合膜快速、便捷地选择性捕获放射性碘的过滤工艺。本报告中的详细操作流程将为纳米材料在环境科学研究领域的应用提供有价值的参考。

方案

1. 柠檬酸盐稳定的金纳米颗粒的合成

  1. 用王水(浓盐酸与浓硝酸按3:1体积比混合)清洗一个双颈圆底烧瓶(250 mL)和一个磁力搅拌子。
    注意:王水溶液具有极强腐蚀性,若操作不慎,可能导致爆炸或皮肤灼伤。
  2. 用去离子水彻底冲洗玻璃器皿,以去除残留的酸液。
  3. 向双颈圆底烧瓶(250 mL)中加入120 mL氯金酸溶液(HAuCl4,1 mM),并在持续搅拌下加热至回流。
  4. 迅速向双颈圆底烧瓶中加入12 mL柠檬酸钠三碱溶液(35 mM),并将所得混合物继续回流20分钟,以确保金盐完全还原。
  5. 让纳米颗粒的胶体悬浮液(深红色)冷却至室温。
  6. 使用石英比色皿(光程1 cm),在520 nm波长下通过紫外-可见分光光度法测定金纳米颗粒(AuNPs)的浓度(消光系数为2.8 × 108)。
  7. 取一滴AuNPs悬浮液滴加到覆碳铜网(400目)上,在室温下干燥,然后通过透射电子显微镜(TEM)测定AuNPs的粒径。
  8. 将胶体金纳米颗粒悬浮液保存于4 °C条件下。

2. 杂化膜(Au-CAM)的制备

  1. 使用注射器装置制备嵌入金纳米颗粒的膜过滤器
    1. 用去离子水(10 mL)清洗由过滤装置支撑的醋酸纤维素膜(孔径:0.45 μm,直径:25 mm),重复三次。
    2. 用无菌注射器(20 mL)吸取10 mL柠檬酸盐稳定的AuNPs(10 nM),缓慢加入预先清洗过的醋酸纤维素膜过滤器中(图1)。
    3. 用10 mL去离子水清洗过滤装置三次,以去除未固定的AuNPs。
      ​注意:固定在醋酸纤维素膜上的AuNPs具有高度稳定性,因此Au-CAM可在常温条件下储存数周,而不会丧失其化学性质或稳定性。
  2. 使用真空泵制备金纳米颗粒膜过滤器
    1. 将醋酸纤维素膜(孔径:0.45 μm,直径:47 mm)置于烧结玻璃支撑的滤器夹具(直径:40 mm)与刻度漏斗(300 mL)之间。
    2. 将烧结玻璃支撑与刻度漏斗的组合单元连接至回收瓶(500 mL)和真空泵。
    3. 向刻度漏斗中加入10 mL柠檬酸盐稳定的AuNPs(10 nM),然后施加真空,直至所有AuNPs完全通过醋酸纤维素膜(约20秒)。
    4. 在膜的另一侧重复相同步骤(步骤2.2.3),以在膜的两侧均固定AuNPs。
    5. 在高性能条件下,使用扫描电子显微镜(SEM)分析Au-CAM表面,加速电压最高达15 kV(图2d)。
      注意:为检测Au-CAM在高盐条件下的纳米颗粒稳定性,将复合膜浸入1.0 M NaCl溶液中2小时,随后进行目视检查以确认Au-CAM的稳定性。

3. 在批处理系统中使用 Au-CAM 吸附放射性碘

  1. 将放射性碘([125I]NaI,2.2 MBq)用 3 mL 纯水、1.0 M NaCl 或 10 nM NaI 稀释,并将每种溶液加入培养皿(直径 50 mm × 高 15 mm)中。
    注意:氧化态的放射性碘可能具有挥发性,必须使用适当的铅屏蔽和铅制容器进行操作。所有放射性化学操作均应在通风良好的活性炭过滤通风橱中进行,并需使用放射性检测器监测实验过程。
  2. 将通过真空过滤制备的 Au-CAM 放入放射性碘溶液中,在室温下轻轻振荡。
  3. 在指定时间点(0、5、10、30、60、120 分钟)从培养皿中取出 10 μL 放射性碘溶液,使用自动 γ 计数器测定该等分试样的放射性。
  4. 120 分钟后用纯化水冲洗 Au-CAM,然后使用自动 γ 计数器测定膜上捕获的放射性量(图 3)。

4. 连续流动条件下放射性碘的脱盐

  1. 使用 Au-CAM 滤膜去除放射性碘阴离子(125I-
    1. 将放射性碘(3.7 MBq)溶解于 50 mL 纯水、1× PBS、1.0 M NaCl、0.1 M NaOH、0.1 M HCl、10 mM CsCl、10 mM SrCl2、合成尿液或海水中。
    2. 用无菌注射器(50 mL)吸取每种溶液 50 mL,并通过注射泵以约 1.5 mL/s 的流速使其流经 Au-CAM 滤膜装置(图 1)。
    3. 将 5 mL 滤液转移至塑料样品瓶中,用于测定溶液中的放射性活度。
    4. 使用自动 γ 计数器测定滤液中残留的放射性活度(图 4)。
  2. Au-CAM 滤膜的可重复使用性测试
    1. 将放射性碘溶解于合成尿液或海水(3.7 MBq/50 mL)中。
    2. 用无菌注射器(50 mL)吸取 50 mL 溶液,并通过注射泵以约 1.5 mL/s 的流速将其注入 Au-CAM 滤膜装置中。
    3. 使用同一个 Au-CAM 滤膜装置,重复相同的过滤操作(步骤 4.2.2)共七次。
    4. 将每次获得的 5 mL 滤液转移至塑料样品瓶中,用于测定溶液中的放射性活度。
    5. 使用自动 γ 计数器测定七份滤液中的放射性活度。

结果

我们已展示了使用柠檬酸盐稳定的金纳米颗粒(AuNPs)和醋酸纤维素膜制备Au-CAM的简单方法(图1a)。通过扫描电镜(SEM)观察Au-CAM表面,结果显示纳米材料稳定地负载在纤维素纳米纤维上(图2)。被固定在膜上的纳米颗粒保持稳定,在使用1.0 M NaCl等水溶液连续冲洗时不会从膜上释放。Au-CAM对碘阴离子的吸附容量约为每1 g AuNPs吸附12.2 μmol(24)。为评估脱盐性能,将通过真空辅助法制备的Au-CAM浸入含有2.2 MBq [125I]NaI的水溶液中(图1b)。孵育30分钟后,纯水和1.0 M NaCl溶液中的大部分放射性碘(>99%)被Au-CAM捕获(图3)。另一方...
