Filament Polymerization

Filament polymerization is the ordered assembly of soluble protein subunits into elongated, fiber-like structures, a process central to cellular organization and biochemistry. It typically begins with nucleation, in which a small stable oligomer forms, followed by subunit addition at filament ends when the concentration of free protein exceeds a critical threshold; nucleotide binding and hydrolysis can regulate assembly and disassembly in systems such as actin and tubulin. This dynamic behavior supports cytoskeletal structure, intracellular transport, cell division, and mechanical force generation. Studying filament polymerization helps clarify molecular regulation, cytoskeletal defects, and strategies for modifying biomolecular assemblies.

Early endosome functions depend on F-actin polymerization. Here, we describe a microscopy-based in vitro assay that reconstitutes the nucleation and polymerization of F-actin on early endosomal membranes in test tubes, thus rendering this complex series of reactions amenable to biochemical and genetic manipulations.
