Flash Oxidation Technology

Flash oxidation technology is a rapid chemical labeling method used to examine the structure, conformation, and interactions of biomolecules. In protein studies, a brief pulse of light photolyzes hydrogen peroxide to generate hydroxyl radicals, which react with solvent-exposed amino acid side chains before the reaction is quenched. The resulting oxidation pattern can be measured by mass spectrometry to create a structural footprint of the protein. This approach helps identify binding interfaces, monitor conformational changes, compare folded and unfolded states, and investigate transient biochemical assemblies that may be difficult to characterize using slower labeling methods.

Photolysis of caged compounds allows the production of rapid and localized increases in the concentration of various physiologically active compounds. Here, we show how to obtain patch-clamp recordings combined with photolysis of caged cAMP or caged Ca for the study of olfactory transduction in dissociated mouse olfactory sensory neurons.

The biarsenical dyes FlAsH and ReAsH bind specifically to tetracysteine motifs in proteins and can selectively label proteins in live cells. Recently this labeling strategy has been used to develop sensors for different protein conformations or oligomeric states.
