Insoluble Chromogenic Biomass

Insoluble chromogenic biomass refers to cellular or microbial biomass associated with colored material that does not dissolve readily in the surrounding medium, making biological activity visible. It commonly forms when cells or enzymes convert a soluble chromogenic substrate into an insoluble colored product that remains on, within, or near the biomass. This retained signal can reveal enzyme activity, identify producing cells, and distinguish active populations from inactive material. In biochemistry, insoluble chromogenic biomass supports qualitative screening and spatial analysis because color remains localized rather than dispersing through a solution, while also providing a basis for comparing activity among samples.

This video demonstrates an assay to screen enzymes effective in degrading complex polysaccharides in plant biomass using insoluble chromogenic biomass or ICB. The enzymes hydrolyze ICB to release soluble dye-linked oligosaccharides, and the enzyme activity is assessed by colorimetric analysis of the reaction product.

A high-throughput assay for enzyme screening is described. This multiplexed ready-to-use assay kit comprises of pre-chosen Chromogenic Polymer Hydrogel (CPH) substrates and complex Insoluble Chromogenic Biomass (ICB) substrates. Target enzymes are polysaccharide degrading endo-enzymes and proteases.

The pretreatment of lignocellulosic biomass with protic low-cost ionic liquids is shown, resulting in a delignified cellulose-rich pulp and a purified lignin. The pulp gives rise to high glucose yields after enzymatic saccharification.
