Methyl Branching Position

Methyl branching position is the location of a methyl substituent along a carbon skeleton, a structural feature that influences the properties and biological behavior of molecules. In biochemistry, shifting the branch closer to or farther from a functional group changes molecular shape, steric accessibility, conformational flexibility, and how effectively the molecule packs or interacts with enzymes and membranes. These effects help explain differences among branched-chain fatty acids, amino acid-derived metabolites, and other naturally occurring compounds. Mapping methyl branching positions supports lipid analysis, structural identification, metabolic pathway studies, and the design of molecules with tailored biochemical or biophysical properties.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.

DNA Methylation: Bisulphite Modification and Analysis

The gold standard for DNA methylation analysis is genomic sequencing of bisulphite converted DNA. This method takes advantage of the increased sensitivity of cytosine compared with 5-methylcytosine (5-MeC) to bisulphite deamination under acidic conditions. Unmethylated cytosines can be distinguished from methylated cytosines after PCR amplification of the target genomic DNA.
