Breast Organoids

Breast organoids are three-dimensional, self-organizing cell cultures that model key structural and functional features of breast tissue in a laboratory setting. In bioengineering, researchers grow mammary epithelial cells within supportive extracellular matrix materials, where cell-cell signaling and matrix interactions guide proliferation, differentiation, and formation of tissue-like structures. These models provide a controllable platform for studying mammary development, tissue organization, disease mechanisms, and responses to therapeutic compounds. Because they can reproduce aspects of normal or diseased breast tissue more closely than conventional two-dimensional cultures, breast organoids support tissue engineering, personalized research, and the development of improved experimental models.

Activation of latent mutations with adenovirus-Cre into the mammary ductal system results in a clinically relevant metastatic breast cancer. Incorporation of a YFP promoter allows tracking of distal metastatic tumor cells. This model is useful to study latent metastasis, anti-tumor immunity, and for designing novel immunotherapies to treat breast cancer.
