Chemical Permeabilization

Chemical permeabilization is a method that temporarily increases the permeability of cell membranes, allowing normally restricted molecules to enter or leave cells. It works by exposing cells to carefully controlled chemicals, such as surfactants or organic solvents, that disturb the lipid bilayer and create transient pathways while limiting irreversible membrane damage. In bioengineering, this approach supports intracellular delivery of proteins, nucleic acids, drugs, and fluorescent probes, as well as cell fixation, staining, and analysis of internal structures. Optimizing chemical concentration, exposure time, and recovery conditions is essential because excessive membrane disruption can reduce cell viability and alter cellular function.
