Collagen Matrix Remodeling

Collagen matrix remodeling is the continuous process by which cells alter the composition, organization, and mechanical properties of collagen-rich extracellular matrices. In bioengineered systems, cells deposit and align new collagen, modify crosslinks, and degrade existing fibers through matrix metalloproteinases and related enzymes, thereby changing matrix architecture and stiffness. This dynamic interaction influences cell adhesion, migration, proliferation, and differentiation. Understanding and controlling collagen remodeling helps researchers design tissue-engineering scaffolds, model wound healing and fibrosis, and evaluate how engineered matrices regulate tissue formation and disease progression.

Cell migration is an important part of human development and life. In order to understand the mechanisms that can alter cell migration, we present a planar gradient diffusion system to investigate chemotaxis in a 3D collagen matrix, which allows one to overcome modern diffusion chamber limitations of existing assays.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.

An experimental technique for the treatment of chondral defects in the rabbit's knee joint is described. The implantation of autologous chondrocytes seeded on a matrix is a well-accepted method for the remodeling and repair of articular cartilage lesions providing satisfying long-term results. Matrix-assisted autologous chondrocyte transplantation (MACT) offers a standardized and clinically established implantation method.
