Desktop Pat System

A desktop PAT (process analytical technology) system is a compact laboratory platform that monitors and helps control bioprocesses, linking experimental measurements with product quality and process performance. It typically combines in-line, on-line, or at-line sensors with data acquisition and analysis software to track critical process parameters and quality attributes during cultivation, fermentation, or other unit operations. In bioengineering, these systems support real-time process understanding, early detection of deviations, and more consistent optimization of laboratory workflows. By connecting small-scale experiments to quality-by-design strategies, desktop PAT can accelerate development and improve the transfer of bioprocesses toward larger-scale manufacturing.

We present our optimized high-throughput nucleofection protocol as an efficient way of transfecting primary human monocyte-derived dendritic cells with either plasmid DNA or siRNA without causing cell maturation. We further provide evidence for successful siRNA silencing of targeted gene RIG-I at both the mRNA and protein levels.
